摘要
目的 观察穴位贴敷对哮喘小鼠皮肤及肺组织的瞬时受体电位香草酸亚型1 (transient receptor potential vanilloid subtype1,TRPV1)表达的影响,探讨穴位贴敷治疗哮喘潜在的作用机制,并为“肺主皮毛”理论提供实验依据。方法 将75只BALB/c小鼠分为空白组、模型组、穴位贴敷组、假贴敷组与地塞米松组,采用屋尘螨(dermatophagoides pteronyssinuus,HDM)滴鼻方法制备小鼠哮喘模型,穴位贴敷组小鼠选取双侧“肺俞”“心俞”“膈俞”进行穴位贴敷,假贴敷组小鼠以凡士林代替中药贴敷相同穴位,地塞米松组小鼠给予0.1 m L/10g地塞米松灌胃。14 d后采用EMKA-WBP肺功能检测系统检测各组小鼠气道阻力,天狼星红染色法观察肺组织胶原纤维增生,HE染色法观察肺、皮肤组织炎性浸润,免疫组化法检测肺、皮肤组织TRPV1的表达情况。结果 与空白组比较,模型组、假贴敷组小鼠气道阻力明显升高,胶原纤维异常增生、肺组织炎性浸润明显,肺组织TRPV1表达明显增加(P<0.05),皮肤组织TRPV1表达与空白组比较差异无统计学意义。与模型组比较,穴位贴敷组和地塞米松组小鼠气道阻力降低,胶原纤维异常增生及炎性浸润情况明显改善,肺组织TRPV1表达显著降低(P<0.05);穴位贴敷组小鼠皮肤组织TRPV1表达明显升高(P<0.05);地塞米松组小鼠皮肤组织TRPV1表达与模型组比较差异无统计学意义。结论 穴位贴敷治疗哮喘的机制与其通过刺激局部皮肤中TRPV1的表达来影响哮喘的进程有关,TRPV1可能为“肺主皮毛”理论的物质基础之一。
Objective To observe the effect of acupoint application on the expression of TRPV1 in the skin and lung tissues of asthmatic mice,explore the potential mechanism of acupoint application in the treatment of asthma,and provide a experimental basis for the theory of"Lung governing fur".Methods A total of 75 BALB/C mice were divided into blank group,model group,acupoint application group,sham application group and dexamethasone group.The mouse asthma model was prepared by nasal drop of dermatophagoides pteronyssinuus(HDM).The"Feishu""Xinshu"and"Geshu"acupoints of mice in the acupoint application group and sham application group were selected.Mice in the sham application group applied vaseline instead of traditional Chinese medicine to the same acupoints.Mice in the dexamethasone group was given 0.1 mL/10 g gavage treatment of dexamethasone.After 14 days treatment,the airway resistance of mice in each group was detected by EMKA-WBP pulmonary function detection system,Sirius red staining was used to observe the proliferation of collagen fibers in lung tissue,HE staining was used to observe the inflammatory infiltration in lung and skin tissue,and immunohistochemistry was used to detect the expression of TRPV1 in lung and skin tissue.Results Compared with blank group,airway resistance,abnormal collagen fiber hyperplasia and inflammatory infiltration of lung tissue were significantly increased in model group and sham application group,and TRPV1 expression in lung tissue was significantly increased(P<0.05),TRPV1 expression in skin tissue showed no statistical difference.Compared with model group,airway resistance was decreased,abnormal collagen fiber hyperplasia and inflammatory infiltration were significantly improved,and TRPV1 expression in lung tissue was decreased(P<0.05)in acupoint application group and dexamethasone group.The expression of TRPV1 in the skin of acupoint application group was increased(P<0.05).There was no significant difference in skin TRPV1 expression between model and dexamethasone group.Conclusion The mechanism of acupoint application in treating asthma may be related to the effect of acupoint application on the progression of asthma by stimulating the expression of TRPV1 in the skin,TRPV1 may be one of the material basis of the theory"Lung governing fur".
