中药有效成分基于Nrf2/ARE信号通路改善脊髓损伤的研究述评

摘要 脊髓损伤(spinal cord injury,SCI)是最具破坏性的中枢性损伤之一,可引起受损部位以下肢体和躯干严重的运动功能障碍,甚至永久性残疾或死亡。核因子E2相关因子2(nuclear factor erythroid-2-related factor 2,Nrf2)在抗炎、抗氧化应激、抑制细胞凋亡、维持线粒体功能等方面发挥重要作用。中药作为天然诱导剂,其中含有诸多抗炎和抗氧化活性成分,是激活Nrf2通路的有效途径。中药及其有效成分调控Nrf2信号通路治疗SCI已成为现代医学研究热点。本文综述了Nrf2信号通路及其关键靶点蛋白与SCI的关系,对调控Nrf2信号通路治疗SCI的中药有效成分及可能的作用机制进行系统总结,以期为靶向激动Nrf2治疗SCI的创新药物研发与应用提供参考。 Spinal cord injury(SCI)is one of the most devastating central injuries,causing severe motor dysfunction in the limbs and trunk below the damaged site,and even permanent disability or death.Nuclear factor erythroid-2-related factor2(Nrf2)plays an important role in anti-inflammation,and anti-oxidative stress,inhibiting cell apoptosis,and maintaining mitochondrial function.As a natural inducer,traditional Chinese medicine contains many anti-inflammatory and antioxidant active components,which is an effective way to activate the Nrf2 pathway.The regulation of the Nrf2 signaling pathway by traditional Chinese medicine and its active components in the treatment of SCI has become a hot spot in modern medical research.This article reviews the relationship between the Nrf2 signaling pathway and its key target proteins and SCI,and systematically summarizes the effective components and possible mechanisms of traditional Chinese medicine regulating the Nrf2 signaling pathway in the treatment of SCI,to provide a reference for the development and application of innovative drugs targeting Nrf2 in the treatment of SCI.
