摘要 目的:基于传统煎药工艺,建立生香附、醋香附和四制香附的定性定量分析方法。方法:依据传统煎药工艺,制备生香附、醋香附和四制香附挥发油和水煎液。采用薄层色谱(TLC)法,以香附烯酮和α-香附酮为对照,对上述三者的挥发油定性鉴别;以香附对照药材为对照,对上述三者水煎液定性鉴别。采用高效液相色谱法,建立生香附、醋香附及四制香附中香附烯酮和α-香附酮含量测定方法。结果:在TLC色谱图中,斑点清晰、分离度好。建立的含量分析法,香附烯酮、α-香附酮分别在0.109~1.088μg,0.044~0.444μg的范围内与峰面积呈良好的线性关系(r=1.000),平均回收率依次为102.4%(RSD=1.8%)和102.1%(RSD=1.6%)。同一批香附中香附烯酮和α-香附酮含量呈现生香附>醋香附>四制香附的规律。结论:本方法为生香附、醋香附和四制香附质量控制提供了参考。 Objective:Based on traditional decocting process,to establish the qualtitative and quantitative analysis methods of raw Cyperi Rhizoma,vinegar-processed Cyperi Rhizoma and Cyperi Rhizoma processed with four excipients.Methods:Using the traditional decocting technology to prepare the volatile oil and water decoction of raw Cyperi Rhizoma,vinegar-processed Cyperi Rhizoma and Cyperi Rhizoma processed with four excipients.Thin layer chromatography(TLC)was used as the qualitative identification method,and the volatile oils was identified with cyperotundone andα-cyperone as the control,and the water decoction was identified with the control material of Cyperi Rhizoma as the control.The content of cyperotundone andα-cyperone in raw Cyperi Rhizoma,vinegar-processed Cyperi Rhizoma and Cyperi Rhizoma processed with four excipients were determined by high performance liquid chromatography(HPLC).Results:In TLC chromatogram,the spots were clear and well separated.The content analysis method established showed that the concentration of cyperotundone andα-cyperone had a good linear relationship with the peak area in the range of 0.109-1.088μg,0.044-0.444μg,respectively.The average recoveries were 102.4%(RSD=1.8%)and 102.1%(RSD=1.6%),respectively.The contents of cyperotundone andα-cyperone in the same batch of Cyperi Rhizoma showed the pattern of raw Cyperi Rhizoma>vinegar-processed Cyperi Rhizoma>Cyperi Rhizoma processed with four excipients.Conclusion:The methods established provide reference for the quality control of raw Cyperi Rhizoma,vinegar-processed Cyperi Rhizoma and Cyperi Rhizoma processed with four excipients.
