黄精茎秆中多糖类成分的精制研究

摘要 目的精制黄精茎秆中的多糖类成分,为黄精茎秆的资源综合利用提供研究基础。方法用乙醇沉淀法制备黄精茎秆粗多糖;以多糖脱色率和多糖保留率为指标,采用0.5%双氧水和2%活性炭进行脱色研究;以蛋白质脱除率和多糖保留率为指标,采用Sevage法和三氯乙酸法对脱色后黄精茎秆多糖进行脱蛋白研究。结果采用乙醇沉淀法制备黄精茎秆粗多糖,粗多糖得率为2.80%,粗多糖中多糖含量为11.33%;0.5%双氧水脱色效果比2%活性炭好,其平均脱色率为78.88%,平均多糖损失率为7.74%;与Sevage法相比,三氯乙酸法蛋白脱除率可达73.12%,多糖损失率9.34%。结论双氧水和三氯乙酸法脱色脱蛋白工艺可行,操作简单方便,可为黄精非药用部位的资源利用和开发提供参考。 Objective To refine the polysaccharides in the stem of Huangjing and provide a research basis for the comprehensive utilization of Huangjing stem resources.Methods Ethanol precipitation method was used to prepare crude polysaccharides from Huangjing stem;Using the decolorization rate and retention rate of polysaccharides as indicators,0.5%hydrogen peroxide and 2%activated carbon were used for decolorization research;With the protein removal rate and polysaccharide retention rate as indicators,the deproteinization of polysaccharide from the decolored rhizome of Polygonatum sibiricum was studied by using the Sevage method and Trichloroacetic acid method.Results The results showed that the crude polysaccharide from Huangjing stem was prepared using ethanol precipitation method,with a yield of 2.80%and a polysaccharide content of 11.33%in the crude polysaccharide;The decolorization effect of 0.5%hydrogen peroxide is better than that of 2%activated carbon,with an average decolorization rate of 78.88%and an average polysaccharide loss rate of 7.74%;Compared with the Sevage method,the protein removal rate of the Trichloroacetic acid method was 73.12%,and the polysaccharide loss rate was 9.34%.Conclusion The decolorization and deproteinization process with hydrogen peroxide and Trichloroacetic acid is feasible,simple and convenient,and can provide a reference for the resource utilization and development of non medicinal parts of Huangjing.
