摘要
羊肚菌(Morchella spp.)成功实现大田栽培后,其栽培过程中产生大量分生孢子而形成菌霜。纯培养条件下羊肚菌分生孢子的产生及萌发成为研究重点。为实现羊肚菌分生孢子的室内获取,通过模拟六妹羊肚菌(M.sextelata)室外栽培条件,从培养容器、培养基组分、覆盖物种类、覆盖厚度及覆盖时间这5个方面研究六妹羊肚菌分生孢子产生的室内条件。结果显示:用六棱形玻璃罐头瓶(底径7.6 cm,口径6.6 cm,高9.8 cm,容积280 mL)为容器,以体积比1︰1的土壤(含水量20%)和PDA为培养基,接种六妹羊肚菌菌丝块20 d后,在菌落表面覆盖厚度为1.5 cm的无菌湿润河沙,可在室内获得分生孢子,每平方厘米分生孢子产量为(2.37±0.63)×10^(6)个。
During cultivation of Morchella spp.in the field,a large number of conidia were produced in the cultivation process to form conidium frost.The production and germination of conidia under axenic culture conditions became the research focus.To achieve indoor production of Morchella conidia,the indoor conditions simulating the temperature and soil conditions of M.sextelata cultivation in the field were studied,including culture container,medium composition,mulch type,mulch thickness and mulch time.The results showed that condia were obtained indoors by using hexagonal glass cans(diameter 7.6 cm,caliber 6.6 cm,height 9.8 cm,volume 280 mL)filled with soil+PDA at a volumetric ratio of 1:1(water content 20%).Twenty days after inoculation of M.sextelata mycelium blocks into the above mentioned glass cans,the medium surface was covered with 1.5 cm thick aseptic moist river sand to allow conidium production,and the yield was(2.37±0.63)×10^(6)per square centimeter.
