香菇子实体中高β-葡聚糖含量多糖提取物制备及体外免疫活性

Preparation and In Vitro Activity of β-Glucan-Rich Polysaccharide Extracts from Lentinula edodes Fruiting Bodies

摘要 通过比较不同菌株香菇(Lentinula edodes)子实体中β-葡聚糖含量,筛选出高β-葡聚糖含量的香菇沪香F2子实体。以沪香F2子实体为材料,利用不同工艺制备多糖提取物LF1(0.9%NaCl提取)、LF2(0.9%NaCl提取后,沉淀用80℃水提取)、LF3(0.9%NaCl提取后,沉淀用80℃水提取,沉淀再用5%NaOH提取)、WE(100℃水提取)和AE(100℃水提取后,沉淀用5%NaOH提取),并测定其β-葡聚糖含量。利用高效液相色谱和阴离子色谱分析多糖提取物的重均和数均分子量及单糖组成,并测定其体外激活Dectin-1受体活性。结果表明:LF3的β-葡聚糖含量最高,为68.26%,且其激活Dectin-1受体的活性也最高。研究结果可为制备高β-葡聚糖含量,且免疫活性高的香菇多糖提取物提供参考。 Content ofβ-glucan varies by variety in fruiting bodies of Lentinula edodes.After comparison ofβ-glucan content in fruiting bodies of different L.edodes cultivars,the cultivar F2 with a highβ-glucan content was selected as the experimental material.Using different extraction methods,different extracts were obtained,including LF1(extracted by 0.9%NaCl),LF2(extracted by 0.9%NaCl and the precipitate further extracted by hot water at 80℃),LF3(extracted by 0.9%NaCl,the derived precipitate extracted by hot water at 80℃,and then extracted by 5%NaOH),WE(extracted by boiling water at 100℃)and AE(extracted by boiling water at 100℃followed by 5%NaOH).These extracts were measured forβ-glucan content,measured for both weight and number average molecular weight,and analyzed for monosaccharide composition by HPLC and anion chromatography.They were also determined for Dectin-1 receptor activation activity in vitro.The results showed that LF3 showed the highestβ-glucan content(68.26%)and the strongest Dectin-1 activation activity in vitro.These results provided a reference for preparing L.edodes polysaccharides with a highβ-glucan content and strong immunological activity.
