Expression and Effects of Centromeric Protein M in Ovarian Clear Cell Carcinoma Tissues

摘要 目的探讨着丝粒蛋白M(centromere protein M,CENPM)在卵巢透明细胞癌(ovarian clear cell carcinoma,OCCC)组织中的表达及对OCCC中ES2细胞株增殖、迁移、凋亡的影响和作用机制。方法采用HE染色观察OCCC组织病理改变;免疫组织化学法检测OCCC组织及癌旁组织CENPM的表达;si-CENPM转染ES2细胞;Western blot检测CENPM蛋白的表达;CCK-8法、细胞划痕实验检测细胞增殖和迁移能力;流式细胞仪评价CENPM对细胞凋亡和周期的影响。结果与癌旁组织相比,OCCC组织中CENPM高表达(P<0.01);转染si-CENPM#B组ES2细胞CENPM蛋白表达水平较si-NC组降低(P<0.01);与Control组和si-NC组比较,敲减CENPM后ES2细胞的增殖、迁移能力下降(P均<0.05);敲减CENPM后ES2细胞凋亡率增高,阻滞ES2细胞的周期在S期(P均<0.05)。结论下调CENPM可抑制人OCCC中ES2细胞株的增殖、迁移并促进细胞凋亡,提示CENPM参与了OCCC的发生发展。 Objective To investigate the expression of centromere protein M(CENPM)in ovarian clear cell carcinoma(OCCC)and the mechanism and effect on proliferation,migration,and apoptosis of ovarian clear cell carcinoma ES2 cell line.Methods HE staining was used to observe the histopathologic changes of OCCC.The expression of CENPM in OCCC and paracancer tissues was detected by immunohistochemistry.ES2 cell was transfected with si-CENPM.The expression of CENPM protein was detected by western blotting.Cell proliferation and migration were detected by CCK-8 assay and cell scratch assay.The effect of CENPM on apoptosis and cell cycle was evaluated by flow cytometry.Results Compared with paracancer tissue,CENPM was highly expressed in OCCC tissue(P<0.01).The expression level of CENPM protein in ES2 cell transfected with si-CENPM was decreased than that in the si-NC group(P<0.01).Compared with control group and si-NC group,the proliferation and migration ability of ES2 cells was significantly decreased after CENPM deletion(P all<0.05).After CENPM knockdown,ES2 cell apoptosis rate increased and ES2 cell cycle was blocked in S phase(P all<0.05).Conclusion Down-regulating CENPM gene can inhibit the proliferation,migration,and apoptosis of human ovarian clear cell carcinoma ES2 cell line,suggesting that CENPM is involved in the development of ovarian clear cell carcinoma.
