健脾益心方对心梗后心衰大鼠心肌损伤及肠道功能的影响

Effect of Jianpi Yixin Decoction on Myocardial Injury and Intestinal Function of Rats with Heart Failure Following Myocardial Infarction

目的 探索经验方健脾益心方对心梗后心衰大鼠的心肌损伤及肠道功能的影响。方法 通过结扎左冠状动脉前降支复制SD大鼠心梗模型,随机分为假手术组、模型组、健脾益心方组。连续灌胃28 d后以心超观察大鼠心脏结构与功能变化。采用HE和Masson染色法观察心肌或结肠组织病理改变,Western blotting检测肠上皮紧密连接蛋白(occludin)、通过16S rDNA基因测序评估粪便样本中的微生物组成。结果 健脾益心方可降低心梗大鼠LVIDs、LV Vols(P <0.01),显著提高LVEF、LVFS(P <0.001)。病理检测显示健脾益心方可改善心肌组织病理改变及心肌纤维化(P <0.05)。肠道菌群检测发现,模型组杜波西氏菌属、拟普雷沃菌属、纺锤链杆属等升高,颤杆菌克属与罗姆布茨菌属降低,健脾益心方可调节这种变化。此外,健脾益心方还可抑制occludin降低,并改善结肠病理变化。结论 健脾益心方可缓解心梗后心衰大鼠心肌损伤,增强心功能,其对缺血心肌的保护作用可能与改善肠道菌群结构、增强肠道屏障功能有关。

Objective:To explore the effect of experiential recipe Jianpi Yixin Decoction on myocardial injury and intestinal function of rats with post-myocardial infarction(MI)heart failure.Methods:The MI model of SD rats was established surgically through proximal left anterior descending coronary artery ligation,and they were ran-domly divided into sham operation group,model group and Jianpi Yixin Decoction group.After 28 days of continu-ous intragastric administration,the cardiac structure and function indexes of rats were observed by echocardiogra-phy.The histopathological changes of myocardium or colon were observed by hematoxylin-eosin(HE)staining and Masson staining.The occludin in intestinal epithelium was detected by Western blotting,and the microbial compo-sition of fecal samples was evaluated by 16S rDNA sequencing.Results:Jianpi Yixin Decoction decreased LVIDs and LV Vols(P<0.01),and significantly increased LVEF and LVFS(P<0.001)in MI rats.Pathological analysis showed that Jianpi Yixin Decoction could improve pathological changes and myocardial fibrosis of MI rats(P<0.05).Results of fecal microbiota detection showed that the intestinal flora of the model group changed like in-creased Dubosiella,Alloprevotella and Fusicatenibacter,but decreased Oscillibacter and Romboutsia.Jianpi Yixin Decoction could regulate these changes.In addition,Jianpi Yixin Decoction could also inhibit the decrease of occlu-din and improve the pathological changes of colon.Conclusion:Jianpi Yixin Decoction alleviates myocardial inju-ry and enhances cardiac function in rats with heart failure after MI.The protective effect of Jianpi Yixin Decoction on ischemic myocardium may be related to improving intestinal microflora structure and enhancing intestinal barri-er function.
