芪仁通脉合剂中枳实的薄层色谱鉴别研究

TLC identification of Fructus aurantii immaturus in Qiren Tongmai mixture

目的 :对芪仁通脉合剂中枳实的薄层色谱鉴别方法进行研究改进。方法 :以橙皮苷为指标成分,将芪仁通脉合剂提取液在层析板上点样,考察影响枳实薄层层析鉴别的因素(供试品制备方法、展开剂系统等),以筛选出最佳的薄层色谱条件。结果:所获得的最佳薄层层析条件为,采用硅胶G薄层板,以三氯甲烷-乙酸乙酯-甲醇-水(3∶8∶4∶2)10℃以下放置的下层溶液为展开剂,以2%三氯化铝乙醇溶液喷雾,晾干后置紫外灯光(365 nm)下检视。色谱斑点清晰,专属性强,且阴性溶液无干扰。结论 :建立的薄层色谱方法简便准确,专属性强,重复性好,可有效提高芪仁通脉合剂的质量标准。

Objective:To study and improve a method for the identification of Fructus aurantii immaturus in Qiren Tongmai mixture by thin layer chromatography(TLC).Methods:With hesperidin as the index component,the extract from Qiren Tongmai mixture was spotted on TLC plate,and the factors affecting their TLC identification such as extraction conditions,developing solvents and so on were investigated so as to screen out the best TLC conditions.Results:The optimal TLC conditions obtained were:silica gel G thin-layer plate,using the lower layer solution of chloroform-ethyl acetate-methanol-water(3:8:4:2)placed below 10℃as the developing solvent,sprayed with 2%aluminum trichloride ethanol solution,dried and inspected under UV light(365 nm).The chromatographic spots were clear with strong specificity and without interference from negative solution.Conclusion:The established TLC method is simple and accurate,with strong specificity and good repeatability,and can effectively improve the quality standard of Qiren Tongmai mixture.
