长链非编码RNA中OIP5-AS1在重度子痫前期孕妇胎盘组织中的表达及意义

摘要 目的探讨长链非编码RNA Opa相互作用蛋白5-反义转录物1(Opa-interacting protein 5 antisense RNA 1,OIP5-AS1)在重度子痫前期孕妇胎盘组织中的表达及其可能作用。方法采用实时定量荧光(RT-qPCR)法测定30例重度子痫前期孕妇和30例正常胎盘中OIP5-AS1和miR-29b表达情况;采用过划痕实验和Transwell侵袭实验检测缺氧条件下向人绒毛膜滋养层细胞系(HTR-8/SVneo细胞)内转染OIP5-AS1对HTR-8/SVneo细胞迁移和侵袭能力的影响;采用RT-qPCR法测定转染OIP5-AS1和OIP5-AS1基因的小干扰RNA(OIP5-AS1 siRNA)后HTR-8/SVneo细胞微小RNA(microRNA,miRNA)miR-29b的表达情况。结果在mRNA水平上,重度子痫前期组胎盘组织中OIP5-AS1较对照组表达降低(P<0.01);过表达OIP5-AS1后,HTR-8/SVneo细胞迁移和侵袭能力增加,差异有统计学意义(P<0.01);过表达OIP5-AS1后HTR-8/SVneo细胞中miR-29b表达下降(P<0.01)而转染OIP5-AS1 siRNA使得miR-29b表达上调(P<0.01)。结论重度子痫前期患者胎盘组织OIP5-AS1表达较低,且可能通过调控miR-29b表达影响滋养细胞生物学功能进而参与子痫前期的发生。 Objective To investigate the expression and possible role of lncRNA OIP5-AS1 in placental tissue of pregnant women with severe preeclampsia.Methods Real-time quantitative fluorescence(RT-qPCR)was used to determine the expression of OIP5-AS1 and miR-29b in 30 pregnant women with severe preeclampsia and 30 normal placentas.The effects of transfection of OIP5-AS1 into human choriotrophoblast cell line(HTR-8/SVneo cells)under hypoxia on the migration and invasion of HTR-8/SVneo cells were examined by scratch assay and Transwell invasion assay.The expression of microRNA(miRNA)miR-29b in HTR-8/SVneo cells after transfection with OIP5-AS1 and OIP5-AS1 gene small interfering RNA(OIP5-AS1 siRNA)was determined by RT-qPCR.Results At the mRNA level,the expression of OIP5-AS1 in severe preeclampsia group(SPE)was lower than that in normal(Normal)group(P<0.01);After overexpression of OIP5-AS1,the ability of cell migration and invasion was increased,and the difference was statistically significant(P<0.01).After OIP5-AS1 overexpression,miR-29b expression decreased in HTR-8/SVneo cells(P<0.01),while transfection with OIP5-AS1 siRNA resulted in up-regulation of miR-29b expression(P<0.01).Conclusions Compared with normal placenta,the expression of OIP5-AS1 in the placenta of patients with severe preeclampsia is lower,and it may affect the biological function of trophoblastic cells by regulating the expression of miR-29b,thus participating in the occurrence of preeclampsia.
