摘要
为揭示甲状腺乳头状癌(Papillary thyroid carcinoma,PTC)的细胞异质性和免疫学特征,使用Seurat包、clusterProfiler包、CellChat包对GSE184362肿瘤及癌旁数据集进行差异基因、富集、细胞通讯分析,并运用HPA数据库对差异表达蛋白进行了验证.结果显示:样本聚类后分成9个主要的细胞亚群,不同样本之间具有明显的细胞异质性;PTC样本具有明显的免疫激活特性、更强的通讯强度;其B细胞具有独特的空间位置,与肿瘤坏死因子活性等细胞通路相关;此外HLA⁃DPA1和S100A4在肿瘤及癌旁中有明显的表达差异.
To reveal the cellular heterogeneity and immunological characteristics of papillary thyroid carcinoma(PTC),GSE184362 tumor and adjacent tumor data sets were analyzed by Seurat package,cluster Profiler package and CellChat package for differential gene,enrichment and cell communication analysis,and HPA database was used to verify the differentially expressed proteins.The results showed that the samples were divided into 9 main cell subsets after clustering,and there was obvious cell heterogeneity among different samples.PTC samples have obvious immune activation characteristics and stronger communication intensity.B cells have a unique spatial location,which is related to tumor necrosis factor activity and other cellular pathways.In addition,the expression of HLA⁃DPA1 and S100A4 was significantly different in tumors and adjacent tumors.
