生长抑制因子4、缺氧诱导因子-1α在宫颈病变组织中的表达及与HR-HPV感染的关系研究

摘要 目的分析生长抑制因子4(ING4)、缺氧诱导因子-1α(HIF-1α)在宫颈病变组织中的表达及与高危型HPV(HR-HPV)感染的关系。方法选取宫颈癌组织25份(宫颈癌组)、宫颈上皮内瘤变组织标本50份(CIN1组、CIN2-3组)、正常宫颈组织标本20份(正常组),术前检测HR-HPV,分析各组ING4、HIF-1α蛋白表达水平及其相关性、与病理参数关系。结果宫颈癌组、CIN1组、CIN2-3组HR-HPV感染率高于正常组(P<0.05);正常组ING4阳性率最高,而HIF-1α阳性率最低,宫颈癌组HIF-1α阳性率最高(P<0.05);感染HR-HPV的组织中,宫颈癌组ING4阳性率最低(P<0.05),而HIF-1α阳性率最高(P<0.05);在感染HR-HPV的宫颈癌及CIN2-3组织中,ING4与HIF-1α水平呈负相关(P<0.05);年龄、淋巴结转移、脉管浸润、肌层浸润深度为感染HR-HPV的宫颈癌患者ING4阴性且HIF-1α阳性的独立危险因素(P<0.05)。结论HR-HPV感染宫颈病变组织中ING4呈低表达,而HIF-1α呈高表达,从而抑制血管内皮细胞增殖、迁移,延缓宫颈病变进展。 Objective To analyze the expression of growth family member 4(ING4)and hypoxia-inducible factor-1α(HIF-1α)in cervical lesions,and its relationship with high-risk HPV(HR-HPV)infection.Methods 25 cervical cancer tissuesamples(cervical cancer group),50 cervical intraepithelial neoplasia(CIN)tissue samples(CIN1 group,CIN2-3 group)and 20 normal cervical tissue samples(normal group)were collected.HR-HPV was detected before surgery.The protein expression levels and correlation of ING4 and HIF-1αin each group,and their relationship with pathological parameters were analyzed.Results The HR-HPV infection rate was higher incervical cancer group,CIN1 group and CIN2-3 group than in the normal group(P<0.05).The ING4 positive rate was the highest,and the HIF-1αpositive rate was the lowest in the normal group.The HIF-1αpositive rate was the highest in the cervical cancer group(P<0.05).In HR-HPV infected tissues,the ING4 positive rate was the lowest,and the HIF-1αpositive rate was the highest in the cervical cancer group(P<0.05).There was a negative correlation between ING4 and HIF-1αlevels in HR-HPV infected cervical cancer and CIN2-3 tissues(P<0.05).Age,lymph node metastasis,vascular invasion,and the depth of myometrial invasion were independent risk factors for ING4 negative and HIF-1αpositive inpatients infected with HR-HPV(P<0.05).Conclusion The low expression of ING4 and high expression of HIF-1a in HR-HPV infected cervical lesions inhibit the proliferation and migration of vascular endothelial cells,thereby delaying the progression of cervical lesions.
