铜藻硫酸化多糖对巨噬细胞RAW264.7的免疫调节作用及其机制 被引量:1

摘要 从铜藻中提取、分离、纯化得到铜藻硫酸化多糖(SHSN1),对SHSN1结构进行表征,研究其免疫调节作用及其机制。结果表明,SHSN1是一个分子量为250 kDa的硫酸化多糖;SHSN1可以促进RAW264.7细胞白介素6(IL-6)、肿瘤坏死因子-α(TNF-α)和白介素10(IL-10)等细胞因子的分泌水平和mRNA表达水平,并且具有剂量依赖性;此外,SHSN1能显著增强免疫信号通路NF-κB信号通路中关键蛋白p-p65及p-IκBα蛋白表达。综上,铜藻硫酸化多糖SHSN1具有一定免疫调节作用,为海洋来源的硫酸化多糖食品和免疫调节剂的开发提供理论支撑,为铜藻的进一步利用和开发提供思路。 A sulfated polysaccharidewas extracted,separated and purified from Sargassum horneri,then the structure of SHSN1 was characterized,and its immunomodulatory effect and mechanism were studied.The results showed that the SHSN1 was a sulfated polysaccharide with a molecular weight of 250 kDa;SHSN1 can increase the secretion and mRNA expression levels of interleukin(IL)-6,tumor necrosis factor-alpha(TNF-α),IL-10 and other immune regulatory factors and in a dose-dependent manner.In addition,SHSN1 significantly enhanced the expression of p-p65 and p-IκBαproteins in the NF-κB signaling pathway.In conclusion,SHSN1 played a certain immunoregulatory effect on RAW264.7 cells,these results provide theoretical support for the development of sulfated polysaccharide food and immunomodulatory agents from marine sources,and provide ideas for the further utilization and development of the medical-edible S.horneri.
