消瘀健脾止鼾方对间歇性缺氧小鼠鼻黏膜组织NF-κB表达的影响

摘要 目的 探讨消瘀健脾止鼾方对间歇性缺氧小鼠鼻黏膜组织NF-κB表达的影响。方法 36只小鼠随机分为三组,每组12只,采用低氧氧仓建立间歇性缺氧小鼠模型,造模成功后给药组给予消瘀健脾止鼾方治疗,干预2周后取材。采用HE染色检测鼻黏膜病理形态,免疫组化检测鼻黏膜NF-κB蛋白表达,PCR检测鼻黏膜NF-κB的mRNA表达。结果 给药组鼻黏膜状态较模型组无明显改变。模型组NF-κB蛋白表达量及mRNA表达量较空白组升高(P<0.05),给药组NF-κB的mRNA表达量较模型组降低(P<0.05)。结论 消瘀健脾止鼾方不能有效改善间歇性缺氧小鼠鼻黏膜病理形态,但可以抑制NF-κB的表达起到抗炎作用。 Objective Exploring the effect of Xiao-Yu-Jian-Pi-Zhi-Han prescription on NF-κB expression in nasal mucosa tissue of mice with intermittent hypoxia.Methods 36 mice were randomly divided into three groups,with 12 mice in each group.Establish an intermittent hypoxia mouse model using a hypoxic oxygen chamber,and after successful modeling,the medication group was treated with Xiao-Yu-Jian-Pi-Zhi-Han prescription.Sampling after 2 weeks of intervention.The nasal mucosa was examined by HE staining,NF-κB protein expression was detected by immunohistochemistry,and NF-κB mRNA expression was detected by PCR in the nasal mucosa.Results The nasal mucosa status was not significantly changed in the drug administration group compared with the model group.The NF-κB protein expression and mRNA expression were higher in the model group compared with the blank group(P<0.05),and the mRNA expression of NF-κB was lower in the drug-administered group compared with the model group(P<0.05).Conclusion Xiao-Yu-Jian-Pi-Zhi-Han prescription cannot effectively improve the pathological morphology of nasal mucosa in intermittently hypoxic mice,but can inhibit the expression of NF-κB to play an anti-inflammatory role.
