摘要
目的通过检测帕金森病(Parkinson's disease,PD)患者血清微小核糖核酸(miRNA,miR)-7和α-突触核蛋白(α-synuclein,α-Syn)水平,分析两者与认知功能障碍的关系。方法收集2020年1月~2022年1月在沧州市中心医院就诊的PD患者106例作为PD组,并根据Hoehn-Yahr分级标准分为六个等级,随机选择同期性别、年龄相匹配的106例健康体检者作为对照组。采用qRT-PCR法检测两组血清miR-7水平,双抗夹心酶联免疫吸附试验(enzyme linked immunosorbent assay,ELISA)法检测血清α-Syn水平;采用简易精神状态量表(mini-mental state examination,MMSE)和蒙特利尔认知评估量表(montrealcognitiveassessment,MoCA)评估PD患者认知功能情况;采用Spearman法分析血清miR-7,α-Syn水平与MMSE评分、MoCA评分的相关性,采用Pearson法分析PD患者血清miR-7与α-Syn水平的相关性。结果PD组血清miR-7(6.95±1.74)水平低于对照组(8.21±1.35),血清α-Syn(6.28±1.05μg/L)水平高于对照组(3.52±0.56μg/L),差异有统计学意义(t=5.890,23.879,均P<0.05)。PD患者Hoehn-Yahr1级、2级、2.5级、3级、4级和5级血清miR-7(8.03±0.99,7.65±0.76,7.12±1.06,6.64±0.65,6.31±0.73,5.93±0.82)水平逐渐降低,α-Syn(3.79±0.39,4.29±0.81,5.48±1.07,6.37±0.82,7.55±1.34,9.01±0.76μg/L)水平逐渐升高,差异均有统计学意义(F=13.201,55.330,均P<0.05)。Spearman法分析显示,PD患者血清miR-7水平和MMSE评分、MoCA评分均呈正相关(r=0.574,0.597,均P<0.05);血清α-Syn水平和MMSE评分、MoCA评分均呈负相关(r=-0.550,-0.537,均P<0.05)。Pearson法分析显示,PD患者血清miR-7与α-Syn水平呈负相关(r=-0.608,P<0.05)。结论随着PD患者病情加重,血清miR-7水平明显降低,α-Syn水平明显升高,且与患者认知功能障碍密切相关。
Objective To detect the levels of serum miRNA(miR)-7 andα-synuclein(α-Syn)in patients with Parkinson’s disease(PD),and analyze the relationship between the two and cognitive dysfunction.Methods 106 patients with PD in Cangzhou Central Hospital from January 2020 to January 2022 were selected as PD group,and they were divided into six grades according to Hoehn-Yahr grading standard,106 healthy people matched with gender and age in the same period were randomly selected as the control group.The qRT-PCR method to detect serum miR-7 levels in the two groups,double resistant sandwich enzyme linked immunosorbent assay(ELISA)method to detect serumα-Syn levels,and the mini-mental state examination(MMSE)and montreal cognitive assessment(MoCA)was used to assess the cognitive function of PD patients.Spearman method was used to analyze the correlation between serum miR-7,α-Syn level and MMSE score,MoCA score,Pearson method was used to analyze the correlation between serum miR-7 andα-Syn level in PD patients.Results The level of serum miR-7(6.95±1.74)in the PD group was lower than that of the control group(8.21±1.35),and the level of serumα-Syn(6.28±1.05μg/L)was higher than that of the control group(3.52±0.56μg/L),the difference was statistically significant(t=5.890,23.879,all P<0.05).The level of serum miR-7(8.03±0.99,7.65±0.76,7.12±1.06,6.64±0.65,6.31±0.73,5.93±0.82)in Hoehn-Yahr grade 1,2,2.5,3,4 and 5 patients with PD gradually decreased,and the level ofα-Syn(3.79±0.39,4.29±0.81,5.48±1.07,6.37±0.82,7.55±1.34,9.01±0.76μg/L)gradually increased,with a statistically significant difference(F=13.201,55.330,all P<0.05).Spearman analysis showed that the serum miR-7 level in PD patients was positively correlated with MMSE score and MoCA score(r=0.574,0.597,all P<0.05),while the serumα-Syn level was negatively correlated with MMSE score and MoCA score(r=-0.550,-0.537,all P<0.05).Pearson analysis showed that there was a negative correlation between serum miR-7 andα-Syn levels in PD patients(r=-0.608,P<0.05).Conclusion With the aggravation of PD patients,the level of serum miR-7 was significantly decreased,and the level ofα-Syn was significantly increased,which was closely related to cognitive dysfunction.
