基于UPLC-Q-Exactive-Orbitrap MS快速鉴定蒲公英根的化学成分

Rapid Identification of Chemical Constituents in Taraxaci Herba Roots Based on UPLC-Q-Exactive-Orbitrap MS

摘要 采用超高效液相色谱-四极杆-静电场轨道阱高分辨质谱(UPLC-Q-Exactive-Orbitrap MS)对蒲公英根70%乙醇提取物中的化学成分进行快速鉴定.色谱柱采用Hypersil GOLD aQ色谱柱(100 mm×2.1 mm,1.9μm),并以含0.1%甲酸的水溶液(A液)和含0.1%甲酸的乙腈(B液)作为流动相.采用以下梯度进行洗脱:0~2 min 5%B液;2~22 min 5%~95%B液;22~27 min 95%B液;27.1~30 min 5%B液.流速为0.3 mL/min,柱温40℃,进样量为5μL.从蒲公英根中鉴定92个化合物,包括6个氨基酸类、17个苯丙素类、3个核苷类、4个黄酮类、4个醛类、2个生物碱类、4个糖类、11个萜类、3个酰胺类、1个香豆素类、23个有机酸类、14个其他类.本研究对蒲公英根的化学成分进行快速鉴定,为进一步研究蒲公英的药效物质基础和资源开发提供依据. Ultra-high performance liquid chromatography-tandem hybrid quadrupole-electrostatic field orbitrap high resolution mass spectrometry(UPLC-Q-Exactive-Orbitrap MS)was used to rapidly identify the chemical constituents in 70%ethanol extract of Taraxaci Herba roots.The type of chromatographic column was Hypersil GOLD aQ column(100 mm×2.1 mm,1.9μm).The mobile phases were 0.1%formic acid-water(liquid A)and 0.1%formic acid-acetonitrile(liquid B)with the elution gradient of 0-2 min 5%liquid B;2-22 min 5%-95%liquid B;22-27 min 95%liquid B;27.1-30 min 5%liquid B.The flow rate was 0.3 mL/min,the column temperature was 40℃,and the injection volume was 5μL.Ninty-two compounds were identified from Taraxaci Herba roots,including six amino acids,seventeen phenylpropanoids,three nucleosides,four flavonoids,four aldehydes,two alkaloids,four carbohydrates,eleven terpenoids,three amides,one coumarin,twenty-three organic acids and fourteen other classes.The chemical constituents in Taraxaci Herba roots were identified in this study,which provides a basis for further study on the pharmacodynamic substance basis and resource utilization of Taraxaci Herba.
