电子加速器辐照对红花粉末及水提液微生物及羟基红花黄色素A含量的影响 被引量:3

Effect of Electron Accelerator Irradiation on Microorganism and Hydroxysafflor Yellow A Content in Powder and Aqueous Extract of Safflower

摘要 为考察电子加速器辐照对红花粉末及水提液中微生物和羟基红花黄色素A含量的影响,分别选用0、4、6、8、10 kGy辐照剂量对红花粉末和红花水提液进行辐照灭菌处理,比较辐照前后红花粉末和红花水提液性状、显微特征、薄层色谱图、主要有效成分羟基红花黄色素A含量,以及细菌总数、霉菌和酵母菌总数及大肠菌群变化。结果表明,不同剂量电子束辐照前后红花粉末和红花水提液性状未见明显变化。与辐照前相比,辐照后红花粉末的显微特征、薄层色谱图以及羟基红花黄色素A含量未见显著改变(P>0.05),辐照后红花水提液的羟基红花黄色素A含量整体存在显著差异(P<0.05),辐照后红花粉末和水提液的细菌总数、霉菌和酵母菌总数均显著下降(P<0.05)。当辐照剂量达到10 kGy时,红花粉末和红花水提液细菌总数、霉菌和酵母菌总数及大肠菌群符合2020版《中华人民共和国药典》规定的标准。综上所述,电子加速器辐照适用于红花粉末的灭菌,但不适用于红花水提液的灭菌。本研究为红花药材、含红花药材及红花提取物的成药辐照灭菌提供了参考。 In order to investigate the effect of electron accelerator irradiation on the number of microorganisms and the content of hydroxysafflor yellow A in safflower powder and safflower aqueous extract,samples were irradiated at different irradiation doses(0,4,6,8,10 kGy).The properties,microscopic characteristics,thin-layer chromatograms,the content of the main effective component hydroxysafflor yellow A,and the changes of the bacteria,fungal yeasts,and coliform groups before and after irradiation were compared.There was no significant change in the properties of safflower powder and safflower aqueous extract before and after irradiation at different doses.Compared with 0 kGy,there was no significant change in the microscopic characteristics,thin-layer chromatography,and the content of hydroxysafflor yellow A of safflower powder after irradiation(P>0.05).However,there was a significant difference in the content of hydroxysafflor yellow A of safflower aqueous and the number of bacterial and fungal yeast colonies in safflower powder and safflower aqueous extract were significantly reduced(P<0.05).When the irradiation dose reaches 10 kGy,the total number of bacteria,fungal yeast,and coliform bacteria in both safflower powder and safflower aqueous extract became up to the 2020 Pharmacopoeia standards.In conclusion,electron accelerator irradiation is suitable for safflower powder sterilization,but not suitable for the sterilization of safflower aqueous extract.This study provided a reference for the irradiation sterilization of safflower medicinal materials,patent medicines containing safflower medicinal materials and safflower extracts.
