刺梨果多糖对黑素瘤A375细胞的作用及其机制 被引量:1

Effects and mechanism of Rose roxburghii Tratt fruits polysaccharide on melanoma A375 cells

摘要 目的:探讨刺梨果多糖对黑色素瘤A375细胞增殖、迁移、侵袭、周期以及凋亡的影响。方法:通过细胞增殖试验、划痕试验、Transwell试验检测刺梨果多糖对A375细胞增殖、迁移、侵袭的作用;流式细胞术、实时荧光定量PCR技术和蛋白质免疫印迹技术检测刺梨果多糖对黑素瘤A375细胞周期和凋亡相关调控因子表达的影响。结果:刺梨果多糖显著抑制黑素瘤A375细胞增殖、迁移和侵袭,且呈时间和剂量正效应;抑制A375细胞CDK-1和CyclinB1的表达,阻滞细胞周期于G2/M期;显著上调凋亡相关蛋白Bax、Caspase-3、Caspase-8、Caspase-9的表达量,下调抗凋亡Bcl-2蛋白的表达,诱导A375细胞凋亡。结论:刺梨果多糖能够抑制黑素瘤A375细胞的增殖并促进细胞凋亡。 OBJECTIVE To investigate the effects of Rose roxburghii Tratt fruits polysaccharides on proliferation,migration,invasion,cell cycle and apoptosis on melanoma A375 cells.METHODS Cell proliferation test,scratch test and Transwell test were used to detect the effects of Rose roxburghii Tratt fruits polysaccharides on A375 cell proliferation,migration and invasion;Flow cytometry,real-time fluorescent quantitative PCR and Western blot were used to detect the effects of Rose roxburghii Tratt fruits polysaccharide on the expression of cell cycle and apoptosis related regulatory factors of melanoma A375 cells.RESULTS Rose roxburghii Tratt fruits polysaccharides significantly inhibited the proliferation,migration and invasion of melanoma A375 cells,with a positive time and dose effect;Rose roxburghii Tratt fruits polysaccharides could inhibit the expression of CDK-1 and CyclinB1 in A375 cells,and block the cell cycle in G2/M phase;Rose roxburghii Tratt fruits polysaccharide significantly up-regulated the expression of apoptosis related proteins Bax,Caspase-3,Caspase-8,Caspase-9,down regulated the expression of anti-apoptotic Bcl-2 protein,and induced A375 cell apoptosis.CONCLUSION Rose roxburghii Tratt fruits polysaccharides can inhibit the proliferation of melanoma A375 cells and promote apoptosis.
