探究中药鹿血晶对骨髓间充质干细胞的增殖作用

A Discussion on Effect of Luxuejing on Proliferation of Bone Marrow Mesenchymal Stem Cells

目的:探究鹿血晶是否通过PI3K/AKT信号通路促进骨髓间充质干细胞(bone marrww-derived mesenchymal stem cells,BMSCs)增殖。方法:通过MTT法检测不同鹿血晶药物浓度对细胞增殖影响,使用DAPI染色检测经药物刺激后的细胞数量,基于MTT法使用细胞周期实验检测细胞生长周期,通过Western blot实验检测PI3K与AKT的蛋白表达。结果:与对照组相比,鹿血晶浓度为200μg/ml时对BMSCs的增殖效果最佳,细胞数量明显增加(P<0.01),在第3天细胞生长进入对数生长期,第5天后开始逐步平稳。与对照组比较,各药物组的PI3K和AKT蛋白的表达均有提升。结论:鹿血晶作用于BMSCs,细胞数量显著增加,并且细胞中PI3K与AKT的蛋白表达提高。所以中药鹿血晶的促进BMSCs增殖作用可能是通过PI3K/AKT信号通路实现的。

Objective:To investigate whether Luxuejing can promote BMSCs proliferation through PI3K/AKT signaling pathway.Methods:MTT assay was used to detect the effect of drug concentration on cell proliferation.DAPI staining was used to detect the number of cells stimulated by drugs,cell cycle assay was used to detect the cell growth cycle based on MTT assay,and the protein expression of PI3K and AKT was detected through Western blot assay.Results:Compared with the control group,20Oμg/ml concentration of Luxuejing had the best effect on the proliferation of BMSCs,and the number of cells increased significantly(P<0.01).The cells entered the logarithmic phase of growthon the 3rd day,and gradually stabilized after the fifth day.Compared with the control group,the expressions of PI3K and AKT were increased in each drug groups.Conclusion:The number of cells and the expression of PI3K and AKT were significantly increased after Luxuejing acted on BMSCs.Therefore,based on PI3K/AKT signaling pathway,Chinese medicine Luxuejing can promote the growth of BMSCs.
