摘要
目的:探讨PRDX6表达水平对食管癌细胞增殖、迁移、侵袭及放射敏感性的影响。方法:应用GEPIA数据库分析食管癌组织PRDX6表达情况;将抑制基因PRDX6表达的“PRDX6-shRNA”质粒转染到EC9706细胞(sh-PRDX6组)。qRT-PCR和Western Blot检测转染效果。CCK8法检测增殖活力。划痕实验检测迁移能力。Transwell实验检测侵袭能力。克隆形成实验检测克隆形成能力。Western Blot检测AKT、p-AKT的蛋白表达量。结果:食管癌组织中PRDX6表达水平明显高于癌旁组织。qRT-PCR和Western Blot证实“PRDX6-shRNA”质粒有效抑制PRDX6基因表达。与对照组相比,sh-PRDX6组的细胞增殖、迁移、侵袭能力显著下降,放射敏感性增强。sh-PRDX6组细胞中AKT的表达水平无明显变化,而p-AKT表达水平明显下降。结论:PRDX6促进食管癌细胞EC9706的增殖、迁移、侵袭及放射抵抗。
Objective:To investigate the effect of PRDX6 expression on the proliferation,migration,invasion and radiosensitivity of esophageal cancer cells EC9706.Methods:The expression of PRDX6 in esophageal cancer tissues was analyzed by GEPIA database."PRDX6-shRNA"plasmids which inhibit the expression of PRDX6 were transfected into EC9706 cells,and were record as sh-PRDX6 group.qRT-PCR and Western Blot were used to detect the transfection effect.CCK8 assay was used to detect the proliferation.Scratch assay was used to detect the migration.Transwell test was used to detect the invasion.Clone formation assay was used to detect the clone formation ability.Western Blot was used to detect the expressions of AKT and p-AKT proteins.Results:The expression of PRDX6 in esophageal carcinoma tissues was significantly higher than that of adjacent tissues.qRT-PCRand Western Blot showed that"PRDX6-shRNA"plasmids effectively inhibited the expression of PRDX6.Compared with control group,cell proliferation,migration and invasion ability of sh-PRDX6 group were significantly reduced,and the radiosensitivity was increased.The expression level of AKT protein in sh-PRDX6 group was not significantly changed,while the expression level of p-AKT protein significantly decreased.Conclusion:PRDX6 promotes the proliferation,migration,invasionand radioresistance of esophageal cancer cells EC9706.
