宫颈癌患者血清miRNA-34a表达水平研究

摘要 目的分析血清miR-34a在宫颈癌中的表达水平,探讨血清miR-34a在宫颈癌中的可能作用。方法收集贵州医科大学附属肿瘤医院2020年10月至2021年3月宫颈癌患者血清为宫颈癌组,同时收集肺癌、结直肠癌患者和健康体检者血清作为疾病对照组和健康对照组,每组20例,提取血清微小RNA,逆转录后,采用实时荧光定量聚合酶链式反应(PCR)检测miR-34a基因的表达水平。结果与健康对照组比较,宫颈癌组miR-34a表达升高(40.2倍,P=0.003)。与肺癌组及结直肠癌组比较,宫颈癌组miR-34a表达明显升高(93.4倍,P<0.001;25.4倍,P=0.005)。结论与健康人群、肺癌及结直肠癌相比,宫颈癌患者血清miR-34a明显升高,提示其可能做为宫颈癌诊断的生物标志来进一步研究;其miR-34a表达水平和肺癌变化趋势一致,与结直肠癌相反,提示miR-34a在不同肿瘤中可能通过不同的机制发挥作用,值得进一步探讨。 【Objective】To analyze the expression level of serum miR-34a in cervical cancer and explore the possible role of serum miR-34a in cervical cancer.【Methods】Serum samples from patients with cervical cancer in Affiliated Tumor Hospital of Guizhou Medical University from October 2020 to March 2021 were collected as the cervical cancer group.Serum samples from patients with lung cancer,colorectal cancer and healthy subjects were also collected as the disease control group and healthy control group,with 20 patients in each group.Serum microRNA was extracted and reverse transcripted.The expression level of miR-34a gene was detected by real-time fluorescence quantitative PCR.【Results】Compared with the healthy control group,the expression of miR-34a was increased in the cervical cancer group,and the difference was statistically significant(40.2 times,P=0.003).Compared with the lung cancer group and the colorectal cancer group,the expression of miR-34a was significantly increased in the cervical cancer group(93.4 and 25.4 times),and the difference was statistically significant(P<0.001 and P=0.005).【Conclusion】Compared with healthy population,lung cancer and colorectal cancer,serum miR-34a was significantly increased in patients with cervical cancer,suggesting that miR-34a may be used as a biomarker for the diagnosis of cervical cancer.Moreover,the expression level of miR-34a was consistent with the variation trend of lung cancer,but contrary to that of colorectal cancer,suggesting that miR-34a may play a role through different mechanisms in different tumors,which is worthy of further investigation.
