摘要
目的观察地黄饮子加减方对APP/PS1双转基因痴呆小鼠行为学及海马突触可塑性相关蛋白的影响,探讨地黄饮子加减方保护海马神经元及改善海马突触可塑性可能作用机制。方法将30只APP/PS1痴呆小鼠随机分为模型组、模型西药组(盐酸多奈哌齐片)、模型中药组(地黄饮子加减方),每组各10只,另将10只C57BL6小鼠设为空白组。Morris水迷宫、旷场实验检测各组小鼠行为学,HE染色、尼氏染色检测各组小鼠海马神经元数量差异,免疫荧光检测各组小鼠GAP43、SYN、PSD-95表达情况。结果与模型组比较,模型中药组小鼠逃避潜伏期时间明显缩短、穿越平台次数增加(P<0.01);与模型组比较,模型中药组小鼠进入中央区次数及中央区活动时间明显增加(P<0.01);与模型组比较,模型中药组小鼠海马神经元损伤情况得到明显改善;与模型组比较,模型中药组小鼠海马突触可塑性相关蛋白GAP43、SYN、PSD-95荧光表达明显增强。结论地黄饮子加减方可改善痴呆小鼠学习记忆能力,改善痴呆小鼠神经元损伤,增强海马神经元突触可塑性相关蛋白表达,从而起到防治痴呆的作用。
Objective To observe the influence of Modified Dihuang Yinzi Decoction on the ethology and hippocampal synaptic plasticity-related proteins in APP/PS1 double transgenic dementia mice,and to explore the possible mechanism of Modified Dihuang Yinzi Decoction on protecting hippocampal neurons and improving hippocampal synaptic plasticity.Methods The 30 APP/PS1 type dementia mice were randomly divided into model group,western medicine model group(Donepezil Hydrochloride Tablets),traditional Chinese medicine model group(Modified Dihuang Yinzi Decoction),10 mice in each group,and another 10 C57BL6 mice were set as blank group.Morris water maze and open field experiments were used to detect the ethology of mice in each group;HE staining and Niselfern staining were employed to detect the difference in the number of hippocampal neurons in each group;and immunofluorescence method was adopted to detect the expression of GAP43,SYN and PSD-95 in each group.Results Compared with the model group,the escape latency time and the number of crossings the platform were significantly shortened among the model mice in the western medicine model group(P<0.01).Compared with the model group,the number of times the mice entered the central region and the activity time of the central region were significantly increased in the TCM model group(P<0.01);the hippocampal neuronal damage of the TCM model group mice was significantly improved;the hippocampal synaptic plasticity-related proteins GAP43,SYN,and PSD-95 fluorescence expression were significantly enhanced in the TCM model group.Conclusions Modified Dihuang Yinzi Decoction can improve the learning and memory abilities of dementia mice and enhance the expression of synaptic plasticity-related proteins in hippocampal neurons,so it plays the role of preventing and treating dementia.
