BCAT1在肝癌中的表达、预后及免疫浸润的生物信息学分析

Bioinformatic analysis of BCAT1 expression,prognosis and immune infiltration in hepatocellular carcinoma

摘要 目的探讨BCAT1在肝癌中的表达及其与预后、免疫浸润的关系。方法TCGA数据库分析多种肿瘤组织中BCAT1 mRNA的表达水平;GEPIA2数据库、HPA数据、TIMER数据库和LinkedOmics数据库分别分析BCAT1 mRNA的表达水平与肝癌患者的生存状态的关系、BCAT1表达定位、BCAT1与肝癌组织中免疫细胞浸润程度的相关性及共表达基因,对共表达基因进行GO、KEGG富集分析,Cytoscape软件鉴定肝癌中与BCAT1共表达基因的Hub基因。结果BCAT1 mRNA在乳腺浸润癌、胆管癌、肝癌等均高表达(P<0.05)。BCAT1在肝癌组织细胞核呈现中等及高表达,在正常组织中呈弱表达或无表达。BCAT1 mRNA表达(高表达)是肝癌患者预后的独立危险因素(P<0.05)。BCAT1高表达的肝癌患者中位生存时间降低(P=0.001)。肝癌组织中BCAT1 mRNA表达水平与B细胞、CD4+T细胞等浸润程度成正相关性(P<0.001)。共表达基因包括APOH、CD86、SYK、AMBP、C3AR1、CYBB、MSR1、FTCD、SERPINC1和PROC。结论BCAT1可作为肝癌患者预后不良的标志物,BCAT1可能通过影响免疫细胞浸润及代谢途径调控肝癌的发生发展。 Objective This paper aims to investigate the BCAT1 expression in hepatocellular carcinoma(HCC)and its rela⁃tionship with the prognosis and the immune infiltration.Methods GEPIA2 database,HPA database,TIMER database and LinkedOmics database were used to analyze the relationship between the expression of BCAT1 mRNA and the survival state of patients with liver cancer,the expression location of BCAT1,the correlation between BCAT1 and the degree of immune cell infil⁃tration in liver cancer tissues and the co-expression genes,respectively.GO and KEGG enrichment analysis were carried out on the co-expressed genes.Cytoscape software was used to identify the Hub gene co-expressed with BCAT1 in HCC.Results BCAT1 mRNA was over-expressed in breast invasive carcinoma,cholangiocarcinoma and hepatocellular carcinoma(P<0.05).BCAT1 expression in HCC tissues was moderate and high,while weakly expressed or not expressed in normal tis⁃sues.The BCAT1 mRNA expression(high expression)was an independent risk factor for prognosis of patients with HCC(P<0.05).The median survival time of HCC patients with high BCAT1 expression was decreased(P=0.001).The correlation analysis showed that the expression of BCAT1 mRNA in HCC tissues was positively correlated with the infiltration degree of B cells,CD4+T cells(P<0.001).APOH,CD86,SYK,AMBP,C3AR1,CYBB,MSR1,FTCD,SERPINC1 and PROC are 10 co-expressed genes of BCAT1 in HCC.Conclusion The expression of BCAT1 is correlated with the poor prognosis of HCC patients.BCAT1 may regulate the hepatocellular carcinoma progression by affecting the infiltration of immune cells and metabolic pathways.
