疏调气机汤水提取工艺正交优化及其对桥本甲状腺炎大鼠治疗作用的研究

摘要 优化疏调气机汤最佳水提取工艺及对桥本甲状腺炎大鼠的治疗作用。以5种成分的总溶出量为指标,通过单因素试验和正交试验法,优选加水量、浸泡时间、煎煮时间等工艺参数;建立桥本甲状腺炎大鼠模型,ELISA法检测大鼠血清中TSH、FT3、IgG4、IFN-γ、TG-Ab、TPO-Ab和TGF-β1水平,HE染色观察大鼠甲状腺病理学形态变化。最佳水提取工艺为加20倍量水,浸泡20 min,煎煮10 min;与模型组比较,疏调气机汤工艺提取物可明显减轻桥本甲状腺炎大鼠的炎性细胞与改善甲状腺滤泡形态,降低血清TSH、IgG4、IFN-γ、TG-Ab、TPO-Ab、TGF-β1和升高FT3水平差异有统计学意义(P<0.05)。优选的疏调气机汤水提取工艺合理、稳定可行,对桥本甲状腺炎大鼠有一定的治疗作用。 To optimize the optimal aqueous extraction process of Shutiaoqiji Decoction and to study the therapeutic effect on rats with Hashimoto thyroiditis.Optimize the process parameters,such as water addition,soaking time and decoction time by single-factor test and orthogonal test method through to the total dissolved amount of 5 compounds as indexes.Detection of TSH,FT3,IgG4,IFN-y,TG-Ab,TPOAb and TGF-β1 levels in rat serum by ELISA and observation of morphological changes in rat thyroid pathology by HE staining.The best extraction process of Shutiaoqiji Decoction was to add 20 times of water,soak for 20 min,and decoct for 10 min.Compared with the model group,the process extract of Shutiaoqiji Decoction could significantly reduced inflammatory cells and improve thyroid follicular morphology in rats with Hashimoto thyroiditis and reduce the level of serum TSH,IgG4,IFN-γ,TG-Ab,TPO-Ab,TGF-β1 and increase the level of FT3 with statistical significance(P<0.05).The preferred extraction process of Shutiaoqiji Decoction was reasonable,stable and feasible,and has a certain therapeutic effecton rats with Hashimoto's thyroiditis.
