摘要
圆二色(CD)光谱是一种广泛使用的测量蛋白质结构和构象变化的方法,可以提供除其他生物物理、化学和结构生物学技术以外的辅助结构信息。为了评价各实验室间CD测量结果的准确性,实施了2022年度高校实验室间比对项目“α-螺旋主导蛋白质的圆二色光谱测定”,项目以一种α-螺旋为主导的蛋白质(细胞色素C)在波长222 nm处的圆二色值作为考核参数,共有来自12个国内高校测试平台和科研院所的实验室参加了此次比对活动。通过基于经典统计法对比对数据的统计分析,表明各参加实验室测量结果偏差均在可接受范围之内,有效验证了不同实验室间进行CD光谱测量的可比性和可靠性。
Circular dichroism(CD)spectroscopy is a widely-used method for examining the structures and conformational changes of proteins,provides complementary structure information to that obtainable by other biophysical,chemical,and structural biology techniques.In order to evaluate the consistency of CD measurement results among laboratories and ensure the accuracy and reliability of the measurement results,in 2022,the inter laboratory comparison project“Measurement of aα-helical predominate protein by circular dichroism spectroscopy”is organized.The project is carried out to compare the CD spectral magnitude of cytochrome C,aα-helical predominate protein,in wavelength of 222 nm.12 test platforms of domestic university and laboratories of scientific research institutes participated in this comparison.Through the effective implementation of the comparison scheme and the statistical calculation of the comparison data based on classical statistic methods,it is found out that the deviation of the measurement results of each participant laboratory is within the acceptable range,which verifies the comparability and reliability of CD spectrum measurement between different laboratories.
