宫颈癌及宫颈癌前病变组织中DEPDC1、miR-216a-5p的表达及其与高危型HPV感染的相关性

Expression of DEPDC1,miR-216a-5p in cervical cancer and cervical precancerous lesions and its correlation with high-risk HPV infection

目的探究DEP结构域蛋白1(DEPDC1)、miR-216a-5p在宫颈癌及宫颈癌前病变组织中的表达及其与高危人乳头瘤病毒(HR-HPV)感染的相关性。方法选取2020年12月至2022年4月汉中市中心医院妇科收治的94例宫颈病变患者为研究对象,其中62例宫颈癌前病变患者的宫颈病变组织作为癌前病变组,32例宫颈癌患者的癌组织作为宫颈癌组。同期选择因患子宫肌瘤行子宫全切患者的30例正常宫颈组织作为对照组。采用q RT-PCR检测DEPDC1、miR-216a-5p的表达水平;采用免疫组化法测定DEPDC1蛋白的表达水平;采用Pearson法分析宫颈组织中DEPDC1与miR-216a-5p表达水平的相关性,以及DEPDC1、miR-216a-5p表达水平与癌前病变组和宫颈癌组HR-HPV感染的相关性。结果与对照组比较,癌前病变组、宫颈癌组DEPDC1的阳性表达率显著升高,差异有统计学意义(P<0.05)。其阳性表达率分别为36.67%、40.32%、100.00%。随着宫颈病变程度的增加,DEPDC1的水平显著升高(1.02±0.21、1.42±0.23、1.78±0.25),miR-216a-5p水平显著降低(1.01±0.11、0.85±0.12、0.68±0.13),HR-HPV感染率显著增加(6.67%、91.94%、100.00%),差异有统计学意义(P<0.05)。经Pearson相关性分析结果显示,癌前病变组、宫颈癌组患者宫颈组织中DEPDC1与miR-216a-5p表达水平呈负相关(r=-0.442,P<0.001)。宫颈组织中DEPDC1表达水平与癌前病变组、宫颈癌组患者HR-HPV感染呈正相关(r=0.296,P=0.004),miR-216a-5p表达水平与癌前病变组、宫颈癌组患者HR-HPV感染呈负相关(r=-0.252,P=0.014)。结论癌前病变和宫颈癌患者宫颈组织中DEPDC的表达水平显著升高,miR-216a-5p水平显著降低,两者与HR-HPV感染有关。

Objective To investigate the expression of DEPDC1,miR-216a-5p in cervical cancer and cervical precancerous lesions and its correlation with high-risk human papillomavirus(HR-HPV)infection.Methods A total of 94 patients with cervical lesions admitted to the Department of Gynecology,Hanzhong Central Hospital from December 2020 to April 2022 were selected as the research objects,among which the cervical lesion tissues of 62 patients with precancerous lesions were taken as the precancerous lesion group,and those of 32 patients with cervical cancer were taken as the cervical cancer group.At the same time,30 patients with hysteromyoma who underwent hysterectomy were selected as the control group.The expression levels of DEPDC1 and miR-216a-5p were detected by qRT-PCR.The expression of DEPDC1 protein was determined by immunohistochemistry.Pearson method was used to analyze the correlation between the expression levels of DEPDC1 and miR-216a-5p in cervical tissues.Spearman method was used to analyze the correlation between the expression levels of DEPDC1,miR-216a-5p and HR-HPV infection in precancerous lesion group and cervical cancer group.Results Compared with the control group,the positive expression rate of DEPDC1 in precancerous lesion group and cervical cancer group was significantly increased(P<0.05),and the positive expression rates were 36.67%,40.32%,and 100.00%,respectively.With the increase of cervical lesion degree,DEPDC1 level was significantly increased:1.02±0.21,1.42±0.23,1.78±0.25;miR-216a-5p level was significantly decreased:1.01±0.11,0.85±0.12,0.68±0.13;HR-HPV infection rate was significantly increased:6.67%,91.94%,100.00%;the differences were statistically significant(P<0.05).Pearson correlation analysis showed that there was a negative correlation between the expression level of DEPDC1 and miR-216a-5p in the cervical tissues of patients in the precancerous lesion group and cervical cancer group(r=-0.442,P<0.001).The expression level of DEPDC1 in cervical tissues was positively correlated with HR-HPV infection in patients in precancerous lesion group and cervical cancer group(r=0.296,P=0.004),and the expression level of miR-216a-5p was negatively correlated with HR-HPV infection(r=-0.252,P=0.014).Conclusion The expression level of DEPDC in cervical tissues of patients with precancerous lesions and cervical cancer is significantly increased,and the level of miR-216a-5p is significantly decreased,both of which are related to HR-HPV infection.
