糙皮侧耳菌株HXS-M1液态发酵中药基质的筛选与优化

Screening and optimization of Chinese herbal matrix for liquid fermentation by Pleurotus ostreatus HXS-M1

试验旨在筛选优化糙皮侧耳菌株HXS-M1液态发酵复方中药的培养基质成分与配比。以糙皮侧耳菌作为复方中药发酵菌种,以菌株产纤维素酶活力为考核指标,采用单因素试验对菌株适宜发酵培养基成分及添加量进行初步筛选;采用响应面Box-Behnken设计建立数学模型,对复方中药液态发酵培养基配比进行优化。结果显示,筛选的发酵中药基质成分为复方中药提取液为碳源、绿豆粉为氮源、碳酸钙为无机盐;优化确定的糙皮侧耳菌液态发酵复方中药培养基质配比为复方中药提取液43.0%、绿豆粉1.0%、碳酸钙0.1%,在此条件下菌株产羧甲基纤维素酶活力最为理想,达到39.92 U/mL。研究表明,试验筛选确定了糙皮侧耳菌HXS-M1液态发酵复方中草药的基质成分与配比,为进一步优选菌株HXS-M1发酵中草药工艺提供了技术参考。

The study was to screen and optimize the fluid-state fermentation medium for compound Chinese matrix by Pleurotus ostreatus strain HXS-M1.Pleurotus ostreatus strain HXS-M1 was used as the fermentation strain,the carboxymethyl cellulase enzyme activity was taken as the response value,the optimal fermentation medium was preliminarily screened by single factor test,and Box-Behneken design in response surface methodology were applied to established mathematical model for optimization of liquid compound Chinese matrix.The results showed that the selected Chinese herbal matrix components were carbon source,Mung bean powder nitrogen source and calcium carbonate inorganic salt,the optimal liquid fermentation medium were 43.0% compound Chinese medicine extract,1.0% Mung bean powder,0.1% calcium carbonate.Under the optimized medium,the cellulase activity was 39.92 U/mL.The study indicates that the composition and proportion of medium for fermentation compound Chinese medicine matrix by Pleurotus ostreatus HXS-M1 are determined,which can provide the technical basis for further optimizing the fermentation process ofl Chinese medicine by strain HXS-M1.
