CD56和Nf在子宫平滑肌瘤中的表达

摘要 目的研究CD56和Nf在子宫平滑肌瘤中的表达及其临床意义。方法收集西安交通大学第二附属医院病理科261例手术切除的子宫平滑肌瘤样本,采用Ventana全自动免疫组化法检测雌激素受体(estrogen receptor,ER)、孕激素受体(progesterone receptor,PR)、平滑肌肌动蛋白(smooth muscle actin,SMA)、结蛋白(Desmin)、神经细胞黏附分子1(neural cell adhesion molecule,NCA1 or CD56)和神经丝(neurofilament,Nf)在261例子宫平滑肌瘤细胞中的表达,并分析上述分子表达的临床意义。同时,以瘤旁非肿瘤性平滑肌作为内对照,以食道平滑肌瘤9例作为外对照;PBS作为阴性对照。结果ER、PR呈胞核型阳性表达,SMA、Desmin、CD56、Nf呈胞质型阳性表达。所有子宫平滑肌瘤均稳定表达ER、PR、SMA、Desmin。CD56在子宫颈平滑肌瘤细胞中的阴性、弱阳性、中等强度阳性和强阳性表达率分别为14.56%(38/261),26.05%(68/261),34.48%(90/261)和24.90%(65/261)。Nf在子宫颈平滑肌瘤细胞中的阴性、弱阳性、中等强度阳性和强阳性表达率分别为44.83%(117/261),30.27%(79/261),18.01%(47/261)和6.90%(18/261)。按照CD56和Nf表达的阳性强度分组,CD56与Nf在子宫平滑肌瘤中的表达无相关性(P<0.05)。同时,SMA、Desmin在食道平滑肌瘤中呈胞质型阳性表达,ER、PR、CD56和Nf在食道平滑肌瘤中呈阴性表达。结论CD56和Nf的阳性表达可能是子宫平滑肌瘤特异性的免疫表型特征。 Objective To study the expression and its clinical significance of CD56 and Nf in uterine leiomyoma(ULM).Methods A total of 261 cases of surgically resected uterine leiomyom specimens were collected from the Department of Pathology,Second Affiliated Hospital of Xi’an Jiaotong University from March 2021 to December 2021.The expression of estrogen receptor(ER),progesterone receptor(PR),smooth muscle actin(SMA),Desmin,CD56 and Nf in 261 cases of uterine leiomyoma was detected by immunostaining with fully automated immunostaining instrument Ventana BenchMark XT(Roche,Switzerland).The clinical significance of the above markers in uterine leiomyoma was analyzed.Non-tumorous smooth muscle in adjacent tumor tissue was used as the internal control,and 9 cases of esophageal leiomyoma samples were used as the external control.PBS was used as the negative control.The clinicopathological features in uterine leiomyomas were analyzed.Results Immunostaining results showed that ER and PR were expressed in nucleus,while SMA,Desmin,CD56 and Nf were expressed in cytoplasm.ER,PR,SMA and Desmin were all stably expressed in uterine leiomyoma.The negative,weakly positive,moderately positive and strongly positive expression rates of CD56 in uterine leiomyoma cells were 14.56%(38/261),26.05%(68/261),34.48%(90/261)and 24.90%(65/261),respectively.The negative,weakly positive,moderately positive and strongly positive expression rates of Nf in uterine leiomyoma cells were 44.83%(117/261),30.27%(79/261),18.01%(47/261)and 6.90%(18/261),respectively.There was no correlation between the expression of CD 56 and Nf in uterine leiomyoma(P<0.05).SMA and Desmin were all stably positively expressed in esophageal leiomyoma samples,and ER,PR,CD56 and Nf were negatively expressed in esophageal leiomyoma samples.Conclusion The positive expression of CD56 and Nf may be a specific immunophenotype in ULM.
