蜡样芽孢杆菌的筛选鉴定及培养优化研究

摘要 试验旨在优化蜡样芽孢杆菌的培养条件。试验从浓香型白酒窖泥中分离筛选获得1株蜡样芽孢杆菌疑似株,进行16S rDNA分析、形态学鉴定、生理生化试验以及生长曲线测定等,采用单因素及响应面分析筛选蜡样芽孢杆菌的最优培养基及培养条件。结果显示,蜡样芽孢杆菌优化后培养基中最佳碳源葡萄糖10.0 g/L、最佳氮源酵母膏12.0g/L、最佳金属离子Ca^(2+)0.5 g/L、最佳转速160 r/min、最适培养温度30℃、最佳接种量3%、最适pH值7,此时蜡样芽孢杆菌生长最快,最大OD_(600 nm)值达到9.6718,比优化前提高约1.72%。研究表明,试验可为新型绿色微生物饲料的发展提供试验参考。 The experiment was to optimize the culture conditions of Bacillus cereus.A suspected Bacillus cereus strain was isolated and screened from the cellar mud of Luz flavor white wine.16S rDNA analysis,morphological identification,physiological and biochemical tests and growth curve determination were conducted.The optimal medium and culture conditions of Bacillus cereus were screened by single factor and response surface analysis.The results showed that the optimal carbon source of Bacillus cereus was 10.0 g/L glucose,the optimal nitrogen source of 12.0 g/L yeast extract,the optimal metal ion of 0.5 g/L Ca^(2+),and the optimal rotational speed of 160 r/min,the optimum culture temperature of 30℃,the optimum inoculation amount of 3%,and the optimum pH value of 7.Under the condition,Bacillus cereus grew fastest,and the maximum OD600 nm value was 9.6718,which was 1.72%higher than that before optimization.The study indicates that the experiment can provide reference for the development of new green microbial feed.
