HPLC法测定甘草党参粗提物中的甘草酸含量

摘要 试验旨在建立液态饲料原料甘草党参粗提物的甘草酸含量检测方法。采用高效液相色谱法(HPLC),以甲醇-0.2mol/L醋酸铵-冰乙酸(67:33:1)为流动相进行等度洗脱,色谱柱为Agilent-C18(4.6 mm×250 mm,5μm),柱温为30℃,流速为1.0 mL/min,检测波长为250 nm。结果显示,甘草酸检测质量浓度在50~1 000 mg/L范围内,甘草酸的线性方程为Y=7 775.2X-10.3,线性关系良好;该方法的回收率为96.09%,相对标准偏差(RSD)为1.13%。研究表明,试验所用方法准确性、重复性高,适用于测定饲料原料甘草党参粗提物的甘草酸含量。 The paper was to establish a method for the determination of glycyrrhizic acid in the crude extract of Codonopsis licorice as liquid feed material.The HPLC was performed on an Agilent-C18 column(4.6 mm×250 mm,5μm)with methanol-0.2 mol/L ammonium acetate and glacial acetic acid(67∶33∶1)as mobile phases.The column temperature was 30℃,the flow rate was 1.0 mL/min,and the detection wavelength was 250 nm.The results showed that the linear equation of glycyrrhizic acid was Y=7775.2X-10.3.When the concentration of glycyrrhizic acid was in the range of 50~1000 mg/L,the recovery was 96.09%,RSD was 1.13%.The study indicates that the method is accurate and reproducible,which is suitable for the determination of glycyrrhizic acid in the crude extract of Codonopsis licorice.
