摘要 目的:探讨lncRNA TP53TG1通过miR-18a/CDC42轴对SW620细胞发展的影响。方法:分析结肠癌组织、癌旁组织、SW620、NCM460细胞中lncRNA TP53TG1、miR-18a和CDC42的表达。检测下调TP53TG1对细胞增殖、侵袭和上皮间质转化(EMT)的影响。采用生物信息学miRanda分析LncRNA TP53TG1作用靶点,检测TP53TG1和miR-18a的相关性。检测下调miR-18a或上调LncRNATP53TG1通过miR-18a对细胞增殖、侵袭和EMT的影响。TargetScan分析miR-18a靶点,检测miR-18a和CDC42之间的关系。分析CDC42 siRNA对细胞增殖、侵袭和EMT的影响。结果:和癌旁正常组织相比,结肠癌组织中lncRNA TP53TG1和CDC42表达上调,miR-18a表达下调;和NCM460细胞相比,SW620细胞中lncRNA TP53TG1和CDC42表达上调,miR-18a表达下调。lncRNA TP53 TG1 siRNA或CDC42 siRNA抑制细胞增殖、侵袭与EMT;lncRNA TP53TG1靶向miR-18a,miR-18a靶向CDC42。miR-18a inhibitor促进细胞增殖、侵袭与EMT;上调LncRNATP53TG1通过miR-18a促进CDC42表达和细胞发展。结论:lncRNA TP53TG1在结肠癌中表达上调且通过miR-18a/CDC42轴促进SW620细胞增殖、侵袭及其EMT。 Objective:To investigate the effects of TP53TG1 on the development of SW620 cells through miR-18a/CDC42.Methods:The differential expression levels of lncRNA TP53TG1,miR-18a and CDC42 in colon cancer tissues,paracancerous normal tissues,SW620 and NCM460 cells were analyzed.The effects of down-regulation of TP53TG1 on proliferation,invasion and EMT of SW620 cells were detected.miRanda was used to analyze the target of lncRNA TP53TG1,and the correlation between TP53TG1 and miR-18a was detected.The effects of down-regulation of miR-18a or up-regulation of lncRNA TP53TG1 on proliferation,invasion and EMT of cells through miR-18a were detected.The target of miR-18a was analyzed by TargetScan,and the relationship between miR-18a and CDC42 was detected.The effects of CDC42 siRNA on proliferation,invasion and EMT of cells were analyzed.Results:Compared with paracancerous normal tissues,the expressions of LncRNA TP53TG1 and CDC42 were upregulated in colon cancer tissues,and the expression of miR-18a was down-regulated;Compared with NCM460 cells,the expressions of lncRNA TP53TG1 and CDC42 were up-regulated,and the expression of miR-18a was down-regulated in cells.LncRNA TP53TG1 siRNA or CDC42 siRNA inhibited the proliferation,invasion and EMT of cells.LncRNA TP53TG1 targets miR-18a and miR-18a targets CDC42.miR-18a inhibitor promoted the proliferation,invasion and EMT of cells;Up-regulation of lncRNA TP53TG1 promoted CDC42 expression and cell development through miR-18a.Conclusion:LncRNA TP53TG1 was up-regulated in colon cancer and promoted proliferation and invasion and EMT of SW620 cells through miR-18a/CDC42 axis.
