摘要
目的探究T细胞17/调节性T细胞(Th17/Treg)相关细胞因子失衡在子痫前期发病机制中的作用。方法收集50例子痫前期孕妇和30例正常孕妇的血清标本,用ELISA法测定血清中白介素-23(IL-23)及白介素-2受体(IL-2R)的浓度;用细胞微球芯片技术(CBA)测定血清中白介素-6(IL-6)及白介素-10(IL-10)的浓度,再选取10例子痫前期孕妇和6例正常孕妇的胎盘及蜕膜组织进行实时荧光PCR(QPCR)及免疫组化法来进一步验证相关细胞因子的表达情况。结果子痫前期孕妇血清中IL-10、IL-2R、IL-23、IL-6的浓度均高于正常孕妇(P<0.05)。QPCR结果显示与正常孕妇相比,子痫前期患者胎盘及蜕膜中IL-23 mRNA表达差异无统计学意义(P>0.05),而子痫前期孕妇胎盘及蜕膜中IL-10 mRNA的表达水平低于正常孕妇,差异有统计学意义(P<0.05),IL-2R mRNA在子痫前期孕妇胎盘及蜕膜中的表达水平高于正常孕妇,差异有统计学意义(P<0.05)。免疫组化结果显示,IL-10在子痫前期孕妇胎盘及蜕膜中的表达均低于正常孕妇,差异有统计学意义(P<0.05),IL-2R及IL-23在子痫前期胎盘中的表达高于正常孕妇,差异有统计学意义(P<0.05),而两组蜕膜中IL-23及IL-2R的表达差异无统计学意义(P>0.05)。结论与正常孕妇相比,IL-10、IL-23及IL-2R的表达水平在子痫前期孕妇中存在差异,这种差异可能与Th17/Treg细胞免疫失衡有关,提示Th17/Treg免疫失衡介导的相关细胞因子改变可能参与了子痫前期的发病过程。
Objective To explore the role of T cell 17/regulatory T cell(Th17/Treg)related cytokine imbalance in the pathogenesis of preeclampsia.Methods Serum samples from 50 preeclampsia pregnant women and 30 normal pregnant women were collected,and the concentration of interleukin-23(IL-23)and interleukin-2 recepter(IL-2R)in serum was tested using ELISA.Additionally,the concentration of interleukin-6(IL-6)and interleukin-10(IL-10)in serum was measured using cell microsphere array(CBA)technology.Then placental and decidual tissues of 10 pregnant women with preeclampsia and 6 normal pregnant women were selected for further verification of the expression of related cytokines by using QPCR and immunohistochemistry.Results The concentrations of IL-10,IL-2R,IL-23 and IL-6 in the serum of preeclampsia pregnant women were higher than those of normal pregnant women(P<0.05).The QPCR results showed that,compared to normal pregnant women,there was no statistically significant diference in the expression of IL-23 mRNA in the placenta and decidua tssues of preeclampsia patients(P>0.05),while the expression level of IL-10 mRNA in the placenta and decidua tissues of preeclampsia patients was lower than that of normal pregnant women,with a statistically significant difference(P<0.05).The expression level of IL-2R mRNA in the placenta and decidua tissues of preeclampsia patients was higher than that of normal pregnant women,with a statistically significant difference(P<0.05).The immunohistochemical results showed that the expression of IL-10 in the placenta and decidua of preeclampsia pregnant women was lower than that of normal pregnant women,with a statistically significant difference(P<0.05).The expression of IL-2R and IL-23 in the placenta of preeclampsia was higher than that of normal pregnant women,with a statistically significant difference(P<0.05).However,there was no statistically significant difference in the expression of IL-23 and IL-2R in the decidua of the two groups(P>0.05).Conclusion Compared with normal pregnant women,there are differences in the expression levels of IL-10,IL-23,and IL-2R in preeclampsia pregnant women.This difference may be related to Th17/Treg cellular immune imbalance,suggesting that Th17/Treg immune imbalance mediated changes in related cytokines may be involved in the pathogenesis of preeclampsia.
