水蓑衣水提物亚急性毒性实验研究 被引量:1

摘要 目的 研究水蓑衣水提物的亚急性毒性,以期为水蓑衣临床使用的安全性评价提供实验依据。方法 将40只雌雄各半的SPF级小鼠随机分为正常对照组,水蓑衣水提物低、中、高剂量组(10、20、40 g/kg),共4组,每组10只。每天灌胃给药1次,隔天称量各组体质量1次,连续21 d给药后,眼眶静脉取血。处死小鼠,取心脏、脾脏、肝脏、肺脏、肾脏、胸腺,测定脏器系数,并进行组织病理学形态观察。检测血清中谷丙转氨酶(alanine aminotransferase,ALT)、谷草转氨酶(aspartate aminotransferase,AST)、乳酸脱氢酶(lactate dehydrogenase,LDH)、肌酸激酶(creatine kinase,CK)的活力,尿素氮(blood urea nitrogen,BUN)及肌酐(creatinine,Cr)的含量。结果 在给药期间,各组小鼠活动状态良好,无明显毒性反应。各组小鼠21 d内的体质量无显著差异,变化趋势基本一致。与正常对照组相比,水蓑衣水提物高剂量组小鼠心脏指数降低(P<0.05);中剂量组小鼠血清中BUN含量降低(P<0.05);各剂量组小鼠的脾脏、肝脏、肺脏、肾脏、胸腺指数以及血清中ALT、AST、LDH、CK活力、Cr含量与正常对照组相比均没有显著性差异。组织切片均未发现明显病理改变。结论 连续21 d灌胃给予小鼠水蓑衣水提物低、中、高剂量(10、20、40 g/kg),与正常对照组相比,其一般状态、体质量、各项血液生化指标及组织病理学形态均未有明显异常。水蓑衣水提物高剂量(40 g/kg,为人临床用量的80倍)对小鼠心脏指数可能有一定的影响。综上所述,可认为水蓑衣在临床常用量下的亚急性毒性较小,临床使用具有较好的安全性。 Objective To study the subacute toxicity of the water extract from Shuisuoyi(Hygrophila Salicifolia Herba),in order to provide experimental basis for the safety evaluation of clinical use of Shuisuoyi(Hygrophila Salicifolia Herba).Methods Forty SPF mice of half male and half female were randomly divided into four groups:the normal control group,the low-dose,medium-dose and high-dose groups(10,20 and 40 g/kg)of Shuisuoyi(Hygrophila Salicifolia Herba),with 10 mice in each group.Intragastric administration was performed once daily,and body weight was measured every two days for 21 consecutive days.Thereafter,the mice were sacrificed to draw orbital blood and harvest the heart,spleen,liver,lung,kidney,and thymus gland for computation of organ coefficients,the histopathology morphology was observed.The activities of glutamic-pyruvic transaminase(ALT),glutamic-oxalacetic transaminase(AST),lactate dehydrogenase(LDH),creatine kinase(CK)and the contents of blood urea nitrogen(BUN),creatinine(Cr)in the serum were detected.Results During the 21-day treatment course,all groups stayed in good condition and showed similar body weight changes,without observable toxic reactions or significant differences in body weight.Compared with the normal control group,the high-dose group exhibited a significant decrease in its cardiac index(P<0.05);the medium-dose group experienced a significant reduction in the serum BUN level(P<0.05);there was no significant differences between the treatment groups and the normal control group in multiple organ coefficients(including spleen,liver,lung,kidney and thymus gland)and serum ALT,AST,LDH,CK activity,and Cr.Tissue sections displayed no evident pathological lesions.Conclusion Administration of three treatment groups(10,20 and 40 g/kg)of Shuisuoyi(Hygrophila Salicifolia Herba)aqueous extract via intragastric route for 21 days has no significant adverse effects on the general health status,body weight,blood biochemical markers,and histopathology of the treated mice compared with the normal control group.The high dose(40 g/kg)of Shuisuoyi(Hygrophila Salicifolia Herba)aqueous extract appears to reduce the cardiac index in mice,and thus studies are needed to further investigate the association between these factors.In conclusion,Shuisuoyi(Hygrophila Salicifolia Herba)aqueous extract,with a low degree of subacute toxicity under clinical usage,is believed to be safe for clinical use.
