复方身痛逐瘀汤对脊神经结扎模型大鼠神经病理性疼痛影响

Effect of Compound Shentong Zhuyu Decoction(复方身痛逐瘀汤)on Neuropathic Pain in Spinal Nerve Ligation Model Rats

目的 观察复方身痛逐瘀汤对脊神经结扎(spinal nerve ligation,SNL)模型大鼠神经病理性疼痛的影响,为今后身痛逐瘀汤治疗神经病理性疼痛提供实验室证据,为以后身痛逐瘀汤的作用机制研究提供实验方法借鉴。方法 将36只雄性SD大鼠随机分为正常组、正常+溶剂组、神经病理性疼痛(neuropathic pain,NP)模型+溶剂组、NP模型+中药溶剂组、假手术+溶剂组、假手术+中药溶剂组。自造模后3 d起,同一实验员对各实验组大鼠实施灌胃给药。同时于造模后第3、5、7、14天对大鼠各项实验指标进行测定和记录,如大鼠的机械痛阈数值、热痛阈数值及大鼠运动功能的评价和分值,造模后第14天选取大鼠L4-L6的背根神经节用于后期进一步研究。结果 造模后第7天、第14天组内比较,与NP模型+溶剂组比较,NP模型+中药溶剂组的机械痛阈与热痛阈均明显上升,差异具有统计学意义(P<0.05);造模后第7天、第14天与NP模型+溶剂组比较,NP模型+中药溶剂组的大鼠运动功能明显改善,差异具有统计学意义(P<0.05)。结论 身痛逐瘀汤能抑制SNL模型大鼠机械痛觉及热痛觉过敏反应,改善SNL模型大鼠运动功能。

Objective To observe the effect of Compound Shentong Zhuyu Decoction(复方身痛逐瘀汤)on neuropathic pain in spinal nerve ligation(SNL)model rats,so as to provide laboratory evidence for its treatment and experimental method reference for further study on its mechanism.Methods Thirty-six male SD rats were randomly divided into the normal group,normal+solvent group,neuropathic pain(NP)model+solvent group,NP model+Chinese medicine solvent group,sham operation+solvent group and sham operation+Chinese medicine solvent group.Three days after modeling,the same experimenter began to gavage the rats in each experimental group.At the same time,the experimental indexes of rats were measured and recorded on the 3rd,5th,7th and 14th days after modeling,such as the mechanical pain threshold value of rats,the thermal pain threshold value and the evaluation and score of locomotor function in rats.On the 14th day after the experiment,the dorsal root ganglia of L4-L6 were selected for further study.Results On the 7th and 14th day of the experiment,compared within the same group,the mechanical and thermal pain thresholds of the NP model+Chinese medicine solvent group were significantly higher than those of the NP model+solvent group(both P<0.05).On the 7th and 14th day of the experiment,compared with the NP model+solvent group,the locomotor function of the NP model+solvent group was significantly improved and the difference was statistically significant(P<0.05).Conclusion Shentong Zhuyu Decoction can inhibit the mechanical pain and thermal hyperalgesia in SNL model rats and improve the locomotor function in SNL model rats.
