苗药追风伞药材质量标准提升研究

摘要 目的 为追风伞药材质量标准的制订提供参考。方法 观察追风伞药材的性状及显微特征,并进行薄层鉴别,同时测定水分、总灰分、酸不溶性灰分和醇溶性浸出物;采用高效液相色谱(HPLC)法测定药材中金丝桃苷的含量。结果 追风伞药材须根呈棕褐色,茎丛生,茎表面褐色,质脆,易折断,中空,节膨大,呈棕褐色;茎顶叶轮生,披针形至线状披针形。该品粉末呈棕褐色,镜下可见花粉粒、纤维束、螺纹导管、表皮细胞、气孔、腺毛、花粉粒等。薄层鉴别结果清晰,专属性强。15批追风伞药材的水分为5.86%~9.90%、总灰分为7.34%~10.56%、酸不溶性灰分为0.38%~3.96%、醇溶性浸出物为11.59%~22.69%。金丝桃苷质量浓度在0.005~0.119 mg/mL(R^(2)=0.999 9)的范围内具有良好的线性关系;精密度、稳定性、重复性试验的相对标准偏差(RSD)值均小于2.0%;平均加样回收率为100.80%,RSD为1.89%;15批追风伞药材中金丝桃苷的含量为0.003%~0.052%。结论 研究对追风伞药材做了性状及显微鉴别,测定了水分、总灰分、酸不溶性灰分、醇溶性浸出物含量,建立了薄层鉴别方法,可为追风伞药材质量标准的制订提供一定参考。 Objective To provide reference for the quality standard establishment of Zhuifengsan(Lysimachia Paridiformis).Methods To observe the characters and microscopic characteristics of the medicinal materials of Zhuifengsan(Lysimachia Paridiformis),and conducting thin layer identification,while determining moisture content,total ash content,acid insoluble ash content,and ethanol-soluble extract.HPLC method was used to determine the content of hyperoside in the medicinal material of Zhuifengsan(Lysimachia Paridiformis).Results The fibrous root of the medicinal material is light yellow to brown,the stem is tufted,the upper part of the stem is brown,brittle,easy to break,hollow stem,and the nodes are expanded and brown,stems and parietal leaves whorled,lanceolate to linear lanceolate.The powder of this product is dark brown,under the microscope,pollen grains,fiber bundles,threaded vessels,epidermal cells,stomata,glandular hairs,pollen grains,etc.can be seen.The results of TLC identification are clear and specific.The moisture content of 15 batches of Zhuifengsan(Lysimachia Paridiformis)was 5.86%-9.90%,the total ash content was 7.34%-10.56%,the acid insoluble ash content was 0.38%-3.96%,and the ethanol-soluble extract was 11.59%-22.69%.The mass concentration of hyperoside has a good linear relationship in the range of 0.005-0.119 mg/mL(R^(2)=0.9999).RSDs of precision test,stability test and repeatability test were all less than 2.0%.The average recoveries were 100.80%,RSDs were 1.89%.The contents of hyperoside in 15 batches of Zhuifengsan(Lysimachia Paridiformis)were 0.003%-0.052%.Conclusion In this study,the properties and microscopic identification of Zhuifengsan(Lysimachia Paridiformis)were carried out,the contents of water,total ash,acid insoluble ash,alcohol soluble extract were determined,and the TLC identification method,which can provide some reference for the formulation of the quality standard of Zhuifengsan(Lysimachia Paridiformis).
