乌头注射液对大鼠肝肾毒性影响的研究

Effect of Wutou Injection(乌头注射液)on Liver and Renal Toxicity in Rats

研究乌头注射液对大鼠体内肝肾功能的影响,进一步探讨乌头注射液对大鼠肝肾毒性作用。方法 将18只大鼠随机分为3组,即空白组(A),单次给药组(B)和连续7 d多次给药组(C),每组6只。适应性喂养1周后,单次给药组(0.65 mg/kg)肌肉注射1 d, 1次/d,连续多次给药组(0.65 mg/kg)肌肉注射7 d, 1次/d,两组于末次给药后禁食12 h,收集尿液,解剖取血,取完整肝脏及肾脏,称取肝脏、肾脏湿重,计算肝脏指数和肾脏指数,检测大鼠血清和尿液中肝肾功能指标。结果 大鼠肾脏指数、总超氧化物歧化酶(SOD)、丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、甲胎蛋白(AFP)、丙二醛(MDA)、血肌酐(CRE)、总蛋白(TP)、胱抑素C(Cys-C)、总胆汁酸(TBA)、α1-微球蛋白(α1-MG)、β2-微球蛋白(β2-MG)生化指标差异具有统计学意义(P<0.05)。结论 乌头注射液对于肝肾功能有影响,长期使用可能对肝肾产生毒性。

Objective To study the effect of Wutou Injection(乌头注射液)on liver and kidney function and further investigate the toxicity effect of liver and kidney in rats.Methods Eighteen rats were randomly divided into 3 groups,namely,blank group(A),single dose group(B)and seven consecutive days of multiple dose group(C),with 6 mice in each group.One week after adaptive feeding,a single administration group(0.65 mg/kg)was given the muscle injection for 1 d,once a day.The continuous administration group(0.65 mg/kg)was given the muscle injection for 7 d,once a day.The two groups fasted for 12 h after the last administration.The urine was collected and the blood was taken out.The liver and kidney were completely taken out.The wet weights of liver and kidney were weighed,liver index and kidney index wer calculated and the liver and kidney function index in serum and urine of the rats were tested.Results The rat kidney index,total superoxide dismutase(SOD),alanine aminotransferase(ALT),aspartate aminotransferase(AST),alpha fetoprotein(AFP),malondialdehyde(MDA),serum creatinine(CRE),total protein(TP),cystatin C(Cys-C),total bile acid(TBA),α1-microglobulin(α1-MG)andβ2-microglobulin(β2-MG)showed significant differences(P<0.05).Conclusion Wutou Injection has an effect on liver and kidney function,and long-term use may cause toxicity to liver and kidney.
