摘要
目的:基于非靶向代谢组学方法分析过敏康对过敏体质人群血清内源性代谢物的影响,从而探究过敏康对于过敏体质的微观调理机制。方法:应用2009年版《中医体质分类判定标准》及本研究的纳入标准,共招募过敏体质人群18人作为研究对象,将过敏康干预前后的血清代谢物借助非靶向代谢组学技术进行对比,采用多元统计分析方法寻找过敏康干预前后的差异代谢物及变化量,并通过代谢数据库对过敏体质潜在的生物标志物进行鉴定分析。结果:过敏康干预前后血清样本差异代谢物有8种。过敏康干预后β-氨基丙腈、吡咯烷、丝氨酸和澳洲茄胺明显上调,Pyro-L-谷氨酰胺基-L-谷氨酰胺、前列腺素F_(1α)、三七皂苷R9和甘油磷酰胆碱明显下调。代谢通路分析显示,关键通路作用于氰基氨基酸代谢(Cyanoamino acid metabolism)和乙醚脂质代谢(Ether lipid metabolism)。结论:过敏康干预后过敏体质人群发生了氨基酸代谢、能量代谢和生物碱代谢等方面的变化,表明过敏康调理过敏体质的机制可能在于改善氮平衡、降低高水平炎症、清除体内自由基、提升免疫力、调控氨基酸代谢和促进凝血功能等方面。
Objective:To analyze the effects of Guominkang on serum endogenous metabolites of allergic subjects according to non-targeted metabolomics method,so as to clarify the micro-conditioning mechanism of Guominkang on allergic population.Methods:A total of 18 subjects with allergic constitution were recruited as experimental research objects by applying the 2009 edition of TCM Constitution Classification Criteria and the inclusion criteria.The serum metabolites before and after medication were compared by non-targeted metabolomics technology and multivariate statistical analysis was used to find the difference metabolites.The amount of change before and after medication,and the potential biomarkers of allergy were identified and analyzed by metabolic database.Results:A total of 8 kinds of serum metabolites were significantly different between the two groups before and after taking Guominkang.The metabolites of beta-aminopropionitrile,pyrrolidine,serine and solasodine were significantly up-regulated,while pyro-L-glutaminyl-L-glutamine,prostaglandin F_(1α),notoginsenoside R9 and glycerophosphocholine were significantly down-regulated after medication.There is a significant difference in two metabolic pathways,namely cyanoamino acid metabolism and ether lipid metabolism.Conclusions:Changes in amino acid metabolism,energy metabolism and alkaloid metabolism occurred in subjects with allergic constitution after taking Guominkang,indicating that the mechanism of Guominkang regulating allergic constitution may be improving nitrogen balance,reducing high levels of inflammation,removing free radicals in the body,enhancing immunity,regulating amino acid metabolism and promoting coagulation function.
