一种禽类动物关节微透析固定装置的设计和应用研究

摘要 目的设计禽类动物关节微透析固定装置,并进行痛风性关节炎鹌鹑模型的微透析采样应用和验证。方法通过对禽类动物生理习性的掌握,创建禽类动物关节微透析固定装置,并通过对实验动物关节液中的尿酸代谢相关指标进行测定和验证。固定鹌鹑使其处于屈膝状态暴露膝关节,将探针埋置关节腔中,探针进口连接推进器,出口插入收集器中,控制透析泵流速为1μL·min^(-1),采用HPLC检测方法对透析液中尿酸浓度进行定量分析。结果成功建立固定鹌鹑并暴露膝关节部位的装置结构,可顺利进行微透析采样并检测到稳定的尿酸含量。与0 d相比较,鹌鹑模型10 d、30 d膝关节液尿酸水平均具有显著差异(P<0.01,P<0.001)。结论禽类动物关节微透析固定装置结构简单、距离调节灵活、适用性强,应用于鹌鹑痛风性关节炎模型时可行性强,可为痛风性关节炎的深入研究奠定基础。 Objective To design the microdialysis fixation device for the joints of poultry,and apply and verify the microdialysis sampling in the quail model of gouty arthritis.Methods By studying the physiological habits of poultry,we designed a microdialysis fixation device for the joints of poultry,measured and verified the related indexes of uric acid metabolism in the joint fluid of quails.The quails were bent to expose the knee joints,and the probe was embedded in the joint cavity.The inlet of the probe was connected with the propeller and the outlet was inserted into the collector.The flow rate of the dialysis pump was controlled set at 1μL·min^(-1).The concentration of uric acid in dialysate was quantitatively analyzed by HPLC.Results The device was successfully established to fix the quail and expose the knee joint,so that microdialysis sampling was conducted and content of uric acid determined smoothly.The changes of the uric acid in the knee cavity of the quail model of gouty arthritis were observed dynamically.Compared with d0,the level of uric acid in the knee fluid at d10 and d30 was significantly different(P<0.01,P<0.001).Conclusion The microdialysis fixation device for poultry joints has simple structure,flexible adjustment and good applicability,laying a foundation for further study of gouty arthritis.
