摘要
目的采用超高效液相色谱-四极杆-静电场轨道阱高分辨质谱(UHPLC-Q-Orbitrap HRMS)技术快速识别和鉴定糖尿康胶囊中的化学成分。方法采用UPLC-Q-Orbitrap HRMS全面采集糖尿康胶囊的MS^(1)和MS^(2)碎片信息,快速鉴定其化学成分。采用Thermo Accucure aQ C18色谱柱(2.1 mm×150 mm,2.6μm)进行分离,以甲醇(A)-0.1%甲酸水溶液(B)为流动相进行梯度洗脱;加热电喷雾离子源(HESI)检测,正、负离子模式同时扫描获得高分辨质谱信息。结果共鉴定出74个化学成分,包括有机酸及其酯类26个(氨基酸类11个、酚酸类8个、脂肪酸类4个和有机酸酯类3个)、生物碱类8个、黄酮类9个、苯乙醇类7个、核苷类5个、二苯乙烯苷类1个、皂苷类3个、香豆素类3个、蒽醌类2个、其他类化合物10个。其中18个化合物通过与对照品进行比较明确识别。结论UPLC-Q-Orbitrap HRMS技术可以快速、全面地鉴定糖尿康胶囊中的化学成分,为提高其质量标准、阐明药效物质基础及临床合理用药提供重要依据。
Objective To rapidly identify the chemical constituents of Tangniaokang capsules by UHPLC-Q-Orbitrap HRMS.Methods UPLC-Q-Orbitrap HRMS was used to comprehensively collect MS^(1) and MS^(2) fragment information of Tangniaokang capsules to identify their chemical compositions.The gradient elution was performed on the Thermo Accucure aQ C18 column(2.1 mm×150 mm,2.6μm)with methanol(A)and water containing 0.1%formic acid(B)as the mobile phase.Positive and negative ion heated electrospray ion source(HESI)modes were used in the simultaneous sanning to obtain high-resolution mass spectrometric information.Results Totally 74 compounds were identified from Tangniaokang capsules,including 26 organic acids and esters(11 amino acids,8 phenolic acids,4 fatty acids and 3 organic esters),8 alkaloids,9 flavonoids,7 phenylethanols,5 nucleosides,1 stilbene glycoside,3 saponins,3 coumarins,2 anthraquinones and 10 other compounds.Among them,18 compounds were easily identified as compared with the reference.Conclusion Chemical components in Tangniaokang capsules are identified rapidly and comprehensively with UPLC-Q-Orbitrap HRMS,which helps improve the quality standards,and elucidate the basis for pharmacodynamic substances.
