大黄素通过NF-kB-iNOS/COX-2信号通路治疗急性胰腺炎的作用机制研究 被引量:1

摘要 目的石研究大黄素对急性胰腺炎的抗炎治疗作用及相关机制。方法半将24只SPF大鼠随机分为假手术组(Sham组)、蔗糖脂肪酸酯处理组(Z组)、模型组(AP组)和大黄素处理组(E组),每组6只。除Sham组外,每一个组中均须使用L-精氨酸制作急性胰腺炎模型,E组于造完模型后1h及6h分别给予大黄素(60mg/kg)腹腔注射治疗;Z组比照E组,腹腔注射等量的4%的蔗糖脂肪酸酯溶液进行处理;造模实验期间每间隔2h观察大鼠的一般情况,AP组造模12h后处死大鼠,E组及Z组末次给药后12h后处死大鼠,ELISA法检测大鼠血清中TNF-α、IL-6、IL-1β、iNOS和COX-2,TBA法检测大鼠血清中SOD、MDA含量;WesternBlot检测大鼠胰腺组织NF-kB蛋白的表达情况;HE染色法检测胰腺及周围组织中的细胞炎症情况。结果与Sham组相比,AP组大鼠一般情况明显变差,血清TNF-α、IL-6及IL-1β指标升高(P<0.05),SOD活性升高,MDA活性降低(P<0.05)COX-2及iNOS显著升高(P<0.05);与AP组相比,大黄素处理组中TNF-α、IL-6及IL-1β指标明显降低(P<0.05),SOD活性升高,MDA活性降低(P<0.05);COX-2及iNOS明显降低(P<0.05);与Sham组比较,AP组大鼠胰腺组织中NF-kB蛋白表达水平显著增加(P<0.05);与AP组相比,E组NF-kB蛋白表达水平明显降低(P<0.05)。结论大黄素可以通过NF-kB-iNOS/COX-2通路来控制胰腺炎症,从而达到治疗胰腺炎的效果。 Objective To study the anti-inflammatory and therapeutic effects of emodin on acute pancreatitis and its related mechanisms.Method 24 SPF rats were randomly divided into sham surgery group(Sham group),sucrose fatty acid ester treatment group(Z group),model group(AP group),and emodin treatment group(E group),with 6 rats in each group.Except for the Sham group,L-arginine was used to create an acute pancreatitis model in each group.Group E received intraperitoneal injection of emodin(60mg/kg)1 hour and 6 hours after the model was established,respectively;Group Z was treated with intraperitoneal injection of an equal amount of 4%sucrose fatty acid ester solution compared to Group E;During the modeling experiment,the general condition of the rats was observed every 2 hours.After 12 hours of modeling in the AP group,the rats were euthanized.After 12 hours of the last administration in the E and Z groups,the rats were euthanized.The serum TNF-α、IL-6、IL-1β、iNOS and COX-2 of the rats was de-tected by ELISA method;TBA method was used to detect the content of SOD and MDA in rat serum;Western Blot detection of the ex-pression of NF-kB protein in rat pancreatic tissue;HE staining was used to detect cellular inflammation in the pancreas and surrounding tissues.Result Compared with the Sham group,the general condition of rats in the AP group significantly deteriorated,with serum TNF-α、IL-6 and IL-1βincreased(P<0.05),SOD activity increased,MDA activity decreased(P<0.05),COX-2 and iNOS signifi-cantly increased(P<0.05);Compared with the AP group,TNF in the emodin treated group-α、IL-6 and IL-1βThe indicators sig-nificantly decreased(P<0.05),SOD activity increased,and MDA activity decreased(P<0.05);COX-2 and iNOS significantly de-creased(P<0.05);Compared with Sham group,The expression level of NF-kB protein in AP group rats pancreatic tissue was signifi-cantly increased(P<0.05);The expression level of NF-kB protein in group E rats pancreatic tissue decreased significantly compared to AP group(P<0.05).Conclusion Emodin can be expressed through NF-kB-iNOS/COX-2 pathway is used to control pancreatitis and achieve the therapeutic effect of treating pancreatitis.
