中央杏仁核生长抑素阳性神经元参与小鼠急性痒和慢性痒的形态学研究 被引量:1

摘要 目的:观察急性痒和慢性痒小鼠中央杏仁核(CeA)内生长抑素(SST)阳性神经元的激活情况,为其参与痒觉信息的调控提供形态学证据。方法:在小鼠颈背部皮内注射组胺(His)或氯喹(CQ)建立急性痒模型;应用二苯基环丙烯酮(DCP)建立接触性皮炎慢性痒模型。进一步结合免疫荧光染色技术分别观察急性和慢性痒小鼠CeA内SST阳性神经元的激活情况。结果:对照组小鼠30 min内抓挠次数为(16.33±5.13)次;His组小鼠30 min内抓挠次数为(110.66±22.00)次;CQ组小鼠30 min内抓挠次数为(120.33±19.50)次。急性痒模型组小鼠抓挠次数显著高于对照组(P<0.05);DCP模型组小鼠搔抓次数明显高于对照组搔抓次数(P<0.05)。免疫荧光染色结果显示在急性痒状态下,对照组CeA内SST/FOS双标神经元占SST阳性神经元的(3.1±0.99)%;His组CeA内SST/FOS双标神经元占SST阳性神经元的(20.23±1.76)%;CQ组CeA内SST/FOS双标神经元占SST阳性神经元的(19.90±2.58)%;在慢性痒状态下,对照组CeA内SST/FOS双标神经元占SST阳性神经元的(3.73±2.05)%;DCP组CeA内SST/FOS双标神经元占SST阳性神经元的(23.93±4.02)%。结论:急性和慢性痒状态下,小鼠CeA内SST阳性神经元激活,为CeA内SST阳性神经元参与痒觉的信息传递提供了形态学依据。 Objective:To observe the activation of somatostatin(SST)positive neurons in the central amygdala(CeA)of mice with chronic itch and those with acute itch,as well as to provide morphological evidence for itch regulation.Methods:The acute itch model was established by intradermal injection of histamine(His)or chloroquine(CQ)on the nape of the neck,while the chronic itch model of contact dermatitis was established using diphenylcycloallone(DCP).Immunofluorescence staining was used to evaluate the activation of SST positive neurons in the CeA of mice.Results:The number of scratches in the control group was(16.33±5.13)in 30 minutes,whereas the number of scratches in the His group and that in the CQ group were(110.66±22.00)and(120.33±19.50),respectively.In comparison with the control group,scratching times of the CQ group,the His group and DCP group were significantly higher(P<0.05).The results showed that SST/FOS double-labeled neurons accounted for(3.1±0.99)%of SST-positive neurons in the CeA of the control group in the acute itch.In His group,SST/FOS double-labeled neurons accounted for(20.23±1.76)%of SST-positive neurons in CeA.In CQ group,SST/FOS double-labeled neurons accounted for(19.90±2.58)%of SST-positive neurons in CeA.In the chronic itch,SST/FOS double-labeled neurons accounted for(3.73±2.05)%of SST-positive neurons in the CeA of the control group.In DCP group,SST/FOS double-labeled neurons accounted for(23.93±4.02)%of SST-positive neurons in CeA.Conclusion:SST positive neurons are activated in the CeA of mice under acute and chronic itch conditions,which provides morphological evidence that SST positive neurons are involved in the information transmission of itch sensation.
