摘要
目的:应用生物信息学方法分析不同腹膜透析时间的腹膜细胞的免疫相关基因(IRGs)表达差异,探索参与腹膜损伤、腹膜纤维化的重要生物标志物。方法:下载基因芯片数据集GSE125498,利用GEO2R数据库分析平台筛选差异表达基因。从ImmPortPortal数据库下载IRGs列表,并筛选出差异表达的IRGs。应用DAVID、STRING生物信息学分析平台,及Cytoscape软件对筛选所得的差异表达IRGs进行生物学注释(GO)、信号通路富集分析(KEGG)及蛋白质-蛋白质相互作用(PPI)网络分析。结果:与20例短期腹膜透析患者相比,在13例长期腹膜透析患者中筛选出差异表达IRGs 36个(|log2FC|>1.0,校正后P<0.05),其中35个上调基因,1个下调基因。GO分析结果显示差异表达IRG的生物学过程主要富集在免疫应答,细胞学组分主要定位于质膜,子功能主要富集在跨膜信号受体活性,KEGG信号通路主要富集在细胞因子-细胞因子受体的相互作用。在PPI网络中应用MCODE及cytohHubba模块筛选出的关键基因为CCR7、CD40LG、IL7R。结论:随着腹膜透析时间的延长,腹膜细胞的免疫应答活跃,相关基因CCR7、CD40LG、IL7R可能是腹损伤、纤维化形成的重要的免疫学标志物。
Objective:To analyze the differentially expressed immune related genes(IRGs)of peritoneal cells in patients with different terms of peritoneal dialysis and to investigate the potential biological markers related to peritoneal injury and peritoneal fibrosis.Methods:Gene chip database GSE125498 was downloaded,and differentially expressed genes were screened by GEO2R.The IRGs list was downloaded from ImmPortPortal database,then the differentially expressed IRGs were screened out.The DAVID,STRING and Cytoscape software were used to perform gene ontology(GO)analysis,KEGG pathway analysis and protein-protein interaction(PPI)network analysis.Results:Compared with 20 patients with short-term peritoneal dialysis,36 IRGs differentially expressed in 13 patients with long-term peritoneal dialysis(|log2FC|>1.0,P<0.05 after correction),of which 35 up-regulated genes and 1 down-regu-lated gene were selected.GO analysis showed that the screened genes of biological process mainly enriched in the immune response.Cytology components were mainly enriched in the plasma membrane.Molecular functions associated with the transmembrane signaling receptor activity.The KEGG pathway is mainly enriched in the cytokine-cytokine receptor interaction.The key genes selected by MCODE and cytohHubba modules in PPI network were CCR7,CD40LG and IL7R.Conclusion:With the prolongation of peritoneal di-alysis time,the immune response of peritoneal cells is active,the related genes CCR7,CD40LG and IL7R may be important biologi-cal markers in the process of peritoneal injury and peritoneal fibrosis.
