摘要
为了探讨紫锥菊提取物与牛至油联合用药对致病菌体外抑菌作用,采用试管二倍稀释法分别测定牛至油和紫锥菊提取物对3种致病菌的体外最低抑菌浓度(MIC)和最小杀菌浓度(MBC),然后以体外最低抑菌浓度为基础,采用棋盘法测定牛至油与紫锥菊提取物联合分级抑菌浓度(FIC)指数。结果表明牛至油对3种致病菌均有极强抑菌作用,牛至油对金黄色葡萄球菌的杀菌效果最强,MIC与MBC均为3.91μg/mL,对大肠杆菌的MIC与MBC均为7.81μg/mL,对链球菌的MIC为31.25μg/mL,MBC为62.50μg/mL。大肠杆菌和金黄色葡萄球菌对紫锥菊提取物中度敏感。紫锥菊提取物对大肠杆菌的MIC为31.25 mg/mL,MBC为62.50 mg/mL,对金黄色葡萄球菌的MIC为62.50 mg/mL,MBC为125.00 mg/mL。牛至油与紫锥菊提取物联合作用于大肠杆菌FIC指数为0.28,两药联合表现为增强作用;牛至油与紫锥菊提取物联合作用于金黄色葡萄球菌的FIC指数为1.00,两药联合表现为相加作用;牛至油与紫锥菊提取物联合作用于链球菌的FIC指数为1.50,两药联合表现为无关作用。综上所述,紫锥菊提取物与牛至油联合用药对大肠杆菌具有较强的协同作用,对金黄色葡萄球菌有相加作用,能显著降低药物的用量。
In order to investigate the antibacterial effect of Echinacea extract and oregano oil in vitro,the minimum antibacterial concentration(MIC)and minimum bactericidal concentration(MBC)of oregano oil and Echinacea extract against three pathogenic bacteria were first determined by tube double dilution method,then the combined graded inhibitory concentration(FIC)index of oregano oil and Echinacea extract based on the minimum inhibitory concentration in vitro was measured.The results showed that oregano oil had a strong antibacterial effect on all three pathogenic bacteria,with the strongest bactericidal effect on S.aureus,both the MIC and MBC were 3.91μg/mL,both MIC and MBC were 7.81μg/mL for E.coli,the MIC was 31.25μg/mL and the MBC was 62.50μg/mL for Streptococcus.Echinacea extract was also moderately sensitive to E.coli and S.aureus,with MIC of 31.25 mg/mL and MBC of 62.50 mg/mL for E.coli,MIC of 62.50 mg/mL and MBC of 125.00 mg/mL for S.aureus.FIC index was 0.28 in the combination of oregano oil and Echinacea extract against E.coli,and the two drugs showed an enhancement effect.FIC index against S.aureus was 1.00,and the two drugs showed an additive effect.FIC index of combined combination on Streptococcus was 1.50,and the combination showed an unrelated effect.In conclusion,the combination of Echinacea extract and oregano oil has a strong synergistic effect on E.coli and an additive effect on S.aureus,and can significantly reduce the dosage of drugs.
