夏芎调神方对缺血性卒中后失眠大鼠的影响及作用机制

摘要 目的观察夏芎调神方对缺血性卒中后失眠大鼠睡眠、神经功能及血清多巴胺(DA)、5-羟色胺(5-HT)、5-羟吲哚乙酸(5-HIAA)水平的影响,探讨夏芎调神方治疗缺血性卒中后失眠的作用机制。方法将60只雄性SD大鼠随机分为夏芎调神方组、模型组、安定组和对照组,每组15只。除对照组外,其他组大鼠制备缺血性卒中后失眠模型。造模成功后,夏芎调神方组给予夏芎调神方水煎剂0.3 g/kg灌胃,安定组给予地西泮片悬液0.2 mg/mL灌胃,对照组和模型组给予等量生理盐水灌胃,均1次/d,连续灌胃14 d。观察各组大鼠的睡眠和神经功能情况,采用ELISA法测定各组大鼠血清DA、5-HT、5-HIAA水平。结果与模型组比较,夏芎调神方组和安定组大鼠的睡眠节律恢复较快,神经功能评分降低但差异均无统计学意义(P均>0.05),血清DA水平均明显降低(P均<0.05),血清5-HT、5-HIAA水平均明显增高(P均<0.05)。夏芎调神方组大鼠的睡眠情况和神经功能评分与安定组比较差异均无统计学意义(P均>0.05),血清DA水平明显低于安定组(P<0.05),血清5-HT、5-HIAA水平均明显高于安定组(P均<0.05)。结论夏芎调神方能够明显改善缺血性卒中后失眠大鼠的睡眠和神经功能,其作用机制可能与降低血清DA水平,促进5-HT、5-HIAA释放有关。 Objective It is to observe the effects of Xia-Xiong-Tiaoshen decoction on sleep,neurological function and serum levels of dopamine(DA),5-hydroxytryptamine(5-HT)and 5-hydroxyindoacetic acid(5-HIAA)in rats with insomnia after ischemic stroke,and to explore the mechanism of this decoction in the treatment of insomnia after ischemic stroke.Methods Sixty male SD rats were randomly divided into Xia-Xiong-Tiaoshen decoction group,model group,diazepam group and control group,with 15 rats in each group.The rats of all groups except for the control group were prepared to establish models of insomnia after ischemic stroke.After successful modeling,the Xia-Xiong-Tiaoshen decoction group was given Xia-Xiong-Tiaoshen decoction 0.3 g/kg by intragastric administration,the diazepam group was given diazepam tablet suspension 0.2 mg/mL by gavage,the control group and the model group were given an equal amount of normal saline by gavage,all once daily,continuously treated for 14 days.The sleep and neurological function recovery of the rats in each group were observed,and the levels of DA,5-HT and 5-HIAA in serum of rats were determined by ELISA.Results After treatment,compared with the model group,the sleep rhythm of the rats in the Xia-Xiong-Tiaoshen decoction group and diazepam group recovered faster,their scores of neurological function were decreased,but the differences were not significant(all P>0.05),the level of serum DA was significantly decreased(P<0.05),that of serum 5-HT and 5-HIAA were significantly increased(P<0.05).The differences in sleep and neurological function scores between the rats in the Xia-Xiong-Tiaoshen decoction group and diazepam group were not statistically significant(all P>0.05),the level of serum DA in the Xia-Xiong-Tiaoshen decoction group was significantly lower,while the levels of serum 5-HT and 5-HIAA were significantly higher than those in the diazepam group(all P<0.05).Conclusion Xia-Xiong-Tiaoshen decoctiona can obviously improve sleep and neurological function of rats with insomnia after ischemic stroke,and its mechanism may be related to inhibiting the expression of serum DA and promoting the release of 5-HT and 5-HIAA.
