沙棘油对黑曲霉线粒体结构及功能的影响

摘要 该文研究沙棘油对黑曲霉线粒体结构和功能的影响,并评估沙棘油对黑曲霉的抗真菌效果。通过以不同的浓度沙棘油处理黑曲霉菌丝体,提取黑曲霉线粒体,并利用透射电子显微镜对线粒体超微结构进行观察;为进一步验证不同浓度沙棘油处理对黑曲霉线粒体功能的影响,测定黑曲霉线粒体的活性氧、丙二醛水平,细胞膜电位变化,三羧酸循环相关酶(琥珀酸脱氢酶和三磷酸腺苷酶)活性变化。结果表明:沙棘油以剂量依赖的方式诱导黑曲霉线粒体结构损伤,经沙棘油处理后的黑曲霉线粒体表现出明显的空泡化、形成异常小泡和线粒体基质结构遭到破坏的迹象,且导致线粒体琥珀酸脱氢酶和三磷酸腺苷酶活性显著降低,线粒体膜电位明显降低,丙二醛和活性氧水平显著升高。综上,沙棘油可以破坏线粒体膜通透性,导致线粒体结构破坏,从而损害线粒体功能,达到抗真菌效果。 The present study sought to examine how sea buckthorn(SBT)oil impacts mitochondrial and overall functionality in Aspergillus niger.A.niger mycelia were treated with different concentrations of SBT oil.The mitochondria were extracted and their ultrastructure observed using transmission electron microscopy.In addition,the mitochondrial function of A.niger treated with different concentrations of SBT oil was assessed.The levels of reactive oxygen species and malondialdehyde,the changes of cell membrane potential,and the activities of the tricarboxylic acid cycle related enzymes,succinate dehydrogenase and adenosine triphosphatase,were measured.SBT oil induced mitochondrial damage in A.niger in a dose-dependent manner.The mitochondria treated with SBT oil showed obvious vacuolization,formation of abnormal vesicles,and the destruction of mitochondrial matrix structure,resulting in altered succinate dehydrogenase and adenosine triphosphatase activities,reduced mitochondrial membrane potential levels,and markedly elevated malondialdehyde and reactive oxygen species levels.Taken together,these findings suggest that SBT oil can damage mitochondrial membrane permeability,leading to mitochondrial structure destruction and impaired mitochondrial function.Thus,SBT oil may have antifungal activity.
