复合凝聚法制备槲皮素微胶囊及其表征 被引量:4

摘要 该试验采用大豆分离蛋白、明胶2种蛋白质分别结合海藻酸钠、阿拉伯胶和果胶3种多糖,利用复合凝聚法制成6种不同壁材包埋槲皮素,分析槲皮素微胶囊的形态、包埋率、热稳定性,并研究包埋率最高的微胶囊的胃肠液释放趋势及贮藏稳定性。结果表明,显微镜下槲皮素被成功包埋,呈椭球形和不规则的棱柱形结构,槲皮素的较好壁材为明胶和阿拉伯胶,产率为(63.76±0.51)%,包埋率为(66.98±0.38)%;傅里叶红外光谱和X射线衍射结果表明,槲皮素成功被微胶囊化,槲皮素由结晶态转变为无定形态且被复合凝聚的壁材所包裹;热重分析表明,明胶-阿拉伯胶槲皮素微胶囊的热稳定性最佳;体外胃肠液模拟消化显示明胶-阿拉伯胶槲皮素微胶囊在胃液环境中能保持结构稳定,在肠液环境中能缓慢释放,8 h胃肠液释放率为(73.39±3.75)%;微胶囊贮藏研究表明,低温能有效缓解明胶-阿拉伯胶槲皮素微胶囊中槲皮素的损失。 Soy protein isolate and gelatin were combined with sodium alginate,gum arabic,and pectin,respectively,to prepare six different wall material combinations for embedding quercetin by complex coacervation.The morphology,embedding rate,and thermal stability of quercetin microcapsules were analyzed,and the release trend in the gastrointestinal fluid and the storage stability of the microcapsules with the highest embedding rate were studied.The results showed that quercetin was successfully embedded under the microscope,and the microcapsules were ellipsoidal or irregular prismatic.Gelatin and gum arabic were better wall materials for embedding quercetin,with a yield of(63.76±0.51)%and an embedding rate of(66.98±0.38)%.The results of Fourier transform infrared spectroscopy and X-ray diffraction indicated that quercetin had been successfully microencapsulated,transitioning from a crystalline state to an amorphous state and being enveloped by the wall materials via complex coacervation.Thermogravimetric analysis showed that the gelatingum arabic quercetin microcapsules had the best thermal stability.The simulated digestion of the microcapsules in the gastrointestinal fluid in vitro showed that the gelatin-gum arabic quercetin microcapsules could keep the structure stable in the gastric fluid environment and release slowly in the intestinal fluid environment.The release rate of quercetin by the microcapsules in the gastrointestinal fluid reached(73.39±3.75)%in 8 h.Low temperatures could effectively alleviate the loss of quercetin in gelatin-gum arabic quercetin microcapsules.
