摘要
目的:探讨食欲素A(Orexin A,OXA)与酒精戒断反应的关系,验证OXA与戒断症状的相关性。方法:通过建立酒精依赖大鼠模型,选取SD雄性大鼠24只,对其进行编号、称重并随机化分为A、B、C三组(每组8只)。A组作为空白对照组普通饮水,B、C组为模型实验组,以含有6%(v/v)酒精水溶液作为唯一水来源,喂养28 d。对照组在第29 d仅在停水后的6 h进行戒断症状评分;B组、C组在第29 d停止供应酒精水溶液而换为普通饮水,并分别于停酒后的第6 h、18 h进行酒精戒断症状评定,评估完成留取3组血液样本保存并离心处理,取上层血浆后使用酶联免疫吸附法(enzyme linked immunosorbent assay, ELISA)测定OXA浓度。结果:B组与C组戒断分数均高于A组,且B组高于C组,差异有统计学意义(P<0.05)。B组OXA水平高于A组和C组,差异有统计学意义(P<0.05);而C组与A组OXA水平比较,差异无统计学意义(P>0.05)。B组与C组OXA水平与戒断反应评分呈正相关(r=0.70,P<0.05),而与大鼠体重变化、酒精摄入量无相关性(P>0.05)。结论:Orexin系统可能在急性酒精戒断反应中发挥重要作用。
Objective:To investigate the relationship between orexin A(OXA)and alcohol withdrawal response and to verify the correlation between OXA and withdrawal symptoms.Methods:By establishing an alcohol-dependent rat model,24 SD male rats were selected,numbered,weighed and randomly divided into three groups:A,B,and C(8 rats in each group).Group A was used as the blank control group,group B and group C were the model experimental groups,and 6%(v/v)alcohol aqueous solution was used as the only water source for 28 days.In the control group,the withdrawal symptoms were scored only at 6 h after water withdrawal on the 29th day.Group B and group C stopped supplying alcohol solution on the 29th day and replaced it with ordinary drinking water.The alcohol withdrawal symptoms were evaluated at 6 h and 18 h after stopping alcohol.The blood samples of the three groups were preserved and centrifuged.The upper plasma was taken and the OXA concentration was determined by enzyme linked immunosorbent assay(ELISA).Results:The withdrawal scores of group B and group C were higher than those of group A,and those in group B were higher than those in group C,the difference was statistically significant(P<0.05).The level of OXA in group B was higher than that in group A and group C,and the difference was statistically significant(P<0.05),while there was no significant difference in OXA level between group C and group A(P>0.05).The level of OXA in group B and group C was positively correlated with the withdrawal reaction score(r=0.70,P<0.05),while there was no correlation with body weight change and alcohol intake in rats(P>0.05).Conclusion:The orexin system may play an important role in the acute alcohol withdrawal response.
