摘要
目的:探讨长链非编码RNA(lncRNA)SNHG1对口腔鳞状细胞癌(OSCC)细胞N-cadherin、MMP-9表达的影响。方法:利用实时荧光定量PCR检测OSCC组织、癌旁组织和OSCC细胞(CAL27、HN6、SCC25)、口腔上皮角质细胞(HOK)中lncRNA SNHG1的表达量;过表达lncRNA SNHG1,将细胞分为对照组(LV-CN组)和实验组(LV-SNHG1组),分别采用CCK-8细胞计数试剂盒和细胞划痕实验检测两组细胞的增殖和迁移能力,实时定量PCR和Western blot检测两组细胞迁移和侵袭相关基因N-cadherin、MMP-9的表达情况。结果:相比于癌旁组织,OSCC组织中lncRNA SNHG1表达含量降低(P<0.01);相比于HOK,CAL27、HN6中lncRNA SNHG1表达含量降低(P<0.01),SCC25中lncRNA SNHG1的表达含量差异无统计学意义(P>0.05);过表达lncRNA SNHG1可抑制OSCC细胞CAL27增殖、迁移的能力(P<0.01),并可抑制N-cadherin、MMP-9蛋白及基因的表达含量(P<0.01)。结论:lncRNA SNHG1在OSCC组织及细胞中低表达,上调lncRNA SNHG1能有效降低OSCC细胞的侵袭和转移。
Objective:To investigate the effect of long non-coding RNA(lncRNA)small nucleolar RNA host gene 1(SNHG1)on the expression of N-cadherin and matrix metalloproteinase-9(MMP-9)in oral squamous cell carcinoma cells.Methods:Quantitative real-time PCR was used to detect the expression of lncRNA SNHG1 in the tissues of oral squamous cell carcinoma,paracancerous tissues,oral squamous cell carcinoma cells(CAL27,HN6,SCC25)and human oral keratinocyte(HOK)cells.LncRNA SNHG1 was overexpressed.Then the cells were divided into control group(LV-CN group)and experimental group(LV-SNHG1 group).Cell Counting Kit-8(CCK8)and cell scratch test were respectively performed to measure the proliferation and migration ability in the cells in both groups.Quantitative real-time PCR and Western blot assay were used to detect the expression of N-cadherin and MMP-9 that were related to cell migration and invasion in two groups.Results:Compared with that in paracancerous tissues,the expression of lncRNA SNHG1 in oral squamous cell carcinoma tissues was significantly reduced(P<0.01).Compared with that in HOK cells,the expression of lncRNA SNHG1 in CAL27 and HN6 was significantly decreased(P<0.01).There was no statistical significance in the expression of lncRNA SNHG1 in SCC25.Overexpression of lncRNA SNHG1 significantly inhibited the proliferation and migration of CAL27 oral squamous cell carcinoma cells(P<0.01),and the expression of N-cadherin and MMP-9 protein and gene(P<0.01).Conclusion:The expression of lncRNA SNHG1 in oral squamous cell carcinoma tissues and cells was significantly reduced.Increase of lncRNA SNHG1 can effectively reduce the invasion and metastasis of oral squamous cell carcinoma cells.
