摘要
目的本研究将通过高内涵成像仪检测红参、麦冬、五味子的总提取物是否影响葡萄糖转运蛋白4(GLUT4)的易位和葡萄糖摄取,并且验证高内涵成像仪是否可以用于检测GLUT4易位。方法L6-GLUT4myc细胞经过给药和胰岛素刺激30 min后,检测GLUT4易位和葡萄糖摄取。结果红参、麦冬、五味子的提取物显著增加了GLUT4-myc的表达,并且同时也增加了葡萄糖的摄取。结论红参、麦冬、五味子的总提取物是通过增加了GLUT4的易位而增加了葡萄糖的代谢。此外本研究也证明L6-GLUT4myc细胞株可以结合高内涵成像系统进行针对刺激GLUT4易位的筛选研究。
Objective The aims of this study are to monitor whether the total extract of red ginseng,Ophiopogon japonicas,Schisandra chinensis will influence GLUT4 translocation by using high-content screening(HCS).Furthermore,to verify GLUT4 translocation can be tested by HCS.Methods GLUT4 translocation and glucose uptake of L6-GLUT4myc cells was detected after stimulation with insulin or the total extracts of three Chinese medicines for 30 min.Results The total extracts of red ginseng,ophiopogon ophiopogon,and schisandra significantly increased GLUT4-myc expression on the cell surface and also glucose uptake.Conclusion This study demonstrated that L6-GLUT4myc cell lines can be combined with HCS to use in screening that how traditional Chinese medicines influence GLUT4 translocation.
