摘要
目的运用网络药理学技术探讨清通袋泡茶治疗阿片类药物所致便秘(Opioid-induced constipation,OIC)的作用机制。方法在中药系统药理学数据库与分析平台(TCMSP)数据库筛选药物的化合物和靶点,并在Cytoscape软件中构建“中药-活性化合物-靶点”网络图。使用GeneCards数据库得到OIC的靶点,与药物靶点形成交集得到靶蛋白。借助STRING数据库构建靶蛋白相互作用(PPI)网络。最后运用R语言进行靶蛋白的GO/KEGG分析。结果得到清通袋泡茶活性化合物43个、对应靶基因共164个,OIC相关疾病靶基因51个,与药物对应靶基因映射筛选出靶蛋白22个。GO功能富集分析得到GO条目1588条,其中生物过程(Biological process,BP)1536条、细胞组分(Cell component,CC)28条、分子功能(Molecular function,MF)24条。KEGG通路富集得到信号通路142条。结论通过对活性化合物、靶点、通路的分析,探讨清通袋泡茶治疗OIC的机制,为进一步药理实验验证提供基础。
Objective To investigate the mechanism of Qingtong teabag in treating opioid-induced constipation(OIC)by using network pharmacology technology.Methods The active ingredients and targets of the main components of herbs were screened on TCMSP database and the network diagram of“herb-active ingredient-target”was constructed on Cytoscape software.GeneCards database was used to obtain the OIC targets,which were intersected with herbs target to obtain a target protein.The STRING database was used to construct a protein-protein interaction(PPI)network.Functional and pathway enrichment(GO/KEGG)analysis were carried out through language R.Results 43 active ingredients and 164 target genes of Qingtong teabag were obtained.51 disease target genes of OIC were screened out and 22 target proteins were obtained by mapping the target genes corresponding to drugs.A total of 1588 GO items,1536 biological processes,28 cellular components and 24 molecular functions were obtained by GO functional enrichment analysis.142 signal pathways were obtained by KEGG pathway enrichment Conclusion The mechanism of Qingtong teabag in treating OIC was analyzed from the aspects of active ingredients,targets and pathways,so as to provide a preliminary basis for further pharmacological experimental verification.
